摘 要:目的本研究主要使用透射电镜观察粉尘螨(Dermatophagoides farinae)体壁及血体腔超微结构。方法取成年粉尘螨20只,先后用2.5%戊二醛及1%锇酸行前后固定,经脱水、包埋、超薄切片、正染色后行透射电镜观察。结果粉尘螨的表皮由上表皮和前表皮构成,其中上表皮进一步分为表皮质层、蜡层和黏质层,前表皮可分为内表皮和外表皮。前表皮结构分层明显,其内可见许多孔道和薄片。粉尘螨血体腔由体壁围绕而成,各组织器官浸浴在血淋巴中,血淋巴内可见大量线粒体、液泡、糖原颗粒以及少量细菌、吞噬细胞等。结论粉尘螨体壁及血体腔超微结构的观察为其形态学研究提供更全面的实验依据。In this study, we observed the ultrastructure of the integument and haemocoele for adult Dermatophagoides farinae with a transmission electron microscope. Twenty mites were first fixed by using 5 % glutaraldehyde and postfixed by u- sing 1 % osmium tetroxide. The TEM observation was conducted after dehydration, embedding, ultrathin section, and positive staining were finished. Results showed that the curricle of D. farinae consisted of epicuticle and procuticle, in which the epieu- ticle is further divided into eutieulin layer, wax layer and cement layer. The procuticle is composed of endocuticle and exocuti- cle, which was laminated obviously and there were many pore cannals and lamellae in it. The haemocoele of Dermatophagoides farinae was surrounded by the integument, various tissues and organs are immersed in it. There were a large number of mito- chondria, vacuoles, glycogen granules, and a few bacterias and phagocytes in the hemolymph. The ultrastructural observations of the integument and the haemocoele provided a more comprehensive experimental basis for the morphology research of D. farinae.
