摘  要:目的:观察荞麦花叶总黄酮对大鼠急性心肌梗塞后心室重构的影响及其可能机制。方法:结扎左冠状动脉前降支建立大鼠急性心肌梗死模型,设假手术组、模型对照组、卡托普利组(10ms/ks)及荞麦花叶总黄酮低、中、高剂量组(100、200、400ms/kg)。造模后连续灌胃给药4W,1次/d。腹腔静脉取血,放射免疫法检测血清中内皮素(ET)、血管紧张素Ⅱ(AngII)、Ⅲ型前胶原氨端肽(PⅢP)、层粘连蛋白(LN)、透明质酸(HA)的含量。取出心脏称重,计算心脏质量指数(HMI)以及左心室质量指数(LV—MI)。硝基四氮唑蓝(NBT)染色法测定心肌梗死面积。结果:与模型对照组比较,荞麦花叶总黄酮在100—400mg/kg剂量范围内可不同程度降低HMI、LVMI值及心肌梗死面积,同时降低血清中ET、AngII、PⅢP、LN、HA的含量。结论:荞麦花叶总黄酮具有抑制大鼠急性心肌梗塞后心室重构的作用,其机制可能与降低血清中ET、AngII、PⅢP、LN、HA的含量相关。Objective: To observe the intervening effect of total flavones of buckwheat flower and leaf (TFBFL) on ventricular remodeling after acute myocardial infarction in rats. Methods: The myocardial infarction model was induced by ligaturing left anterior descending of coronary artery in rats. The rats were randomly divided into sham group, myocardial infaction model contrast group, TFBFL groups with different doses (100,200,400mg/kg) and captopril group ( 10 mg/kg). The rats in each group were given by ig. for 4 w, once per day. The levels of endothelin(ET), angiotensin II (Ang II), Type III procollagen amino terminal peptide (PIIIP), laminin (LN), hyaluronic acid(HA) were determined by radioimmunoassay. The heart was weighed and calculated heart mass index (HMI) and left venticular mass index (LVMI). The myocardial infarction area was determined by NBT dye method. Results: Compared with the model contrast group, TFBFL at a doserange 100-400mg/kg could decline HMI, LVMI and the myocardial infarction area in various degree, and decrease the levels of ET, Ang II , P III P, LN, HA in serum( P 〈 0.05, P 〈 0.01 ) . Conclusion: TFBFL significantly inhibited ventricular remodeling after acute myocardial infarction in rats, which may be related to decrease the levels of ET, Ang II, P III P, LN, HA in serum.
