摘  要:为了解石河子垦区的CDV流行株H基因遗传变异特点,设计2对特异性引物,通过RTPCR方法对29份临床疑似犬瘟热的病料进行检测,对于检测CDV阳性的病料进行流行株H基因克隆、测序和序列分析,结果检测出7份阳性病料。序列分析发现,7个流行株H基因之间的同源率在91.0%~98.9%,与疫苗株(FJ461699.1)的同源率为86.9%~91.7%,存在一定的变异。糖基化位点及抗原表位预测分析发现,与参考株Onderstepoort相比,7个流行株推导的H蛋白糖基化位点、抗原表位均有不同程度的改变。遗传进化分析显示,CDVshzCDVshz6亲缘关系较近,它们与CDVshz7相距较远。研究结果提示,石河子垦区发病犬中存在cDV变异株。In order to understand the genetic variation of Canine distemper virus (CDV) in the Shihezi area, 29 clinically suspected infection CDV samples were detected by RT-PCR method with two pairs of specific primers designed on H gene, and positive samples were conducted cloning, sequencing and sequence analysis. The results showed that seven positive samples were detected. The sequence of H gene in these seven epidemic strains, with the homologous rate range from 91.0% to 98.9% ,had the homologous rate ranging from 86.9%to 91.7% with the vaccine isolate strains (FJ461699.1), and there were a certain variation. Potential glycosylation sites and antigenic epitopes of the epidemic strains H genes analysis results indicated the seven epidemic strains H genes varied on glycosylation sites, antigenic epitopes compared with the reference strain Onderstepoort. Phylogenetic analysis showed that CDVshzl to CDVshz6 with a close relationship were far from CDVshzT. The results sug gested that there are CDV variant strains existed in diseased dogs in Shihezi area.
