摘 要:目的 探讨 VEGF高表达的卵巢癌中 VEGF基因结构有无异常 .方法 用 RT- PCR方法自胎盘中克隆 VEGFc DNA以作为杂交探针 ;收集临床卵巢癌手术切除标本 ,以正常卵巢组织为对照 ,从中提取基因组 DNA并进行 Southern印迹杂交 .结果 序列分析表明所获得基因正确 ,Southernblot分析 10 / 19呈现出大于 2 3 kb片段和 4.3 kb片段的异常片段 ,而不同于正常对照组织的大于 30 kb,6 .5 kb和 2 .5 kb.AIM To study whether there is a difference in the structure of V EGF gene between the normal tissue and the ovarian cancer overexpressing VEGF. METHODS VEGF cDNA was cloned from placenta by RT PCR. The ovarian cancer samples were collected in clinic.With the normal ovariantissue as the control, genomic DNA from the samples was extracted was extracted and compared with the normal ovarian tissue and Southern blotting was done. RESULTS VEGF cDNA clone from placenta was proved to be correct. There are three fragments (>30 kb,6.5 kb,2.5 kb) in normal control,while 10/19 samples only had two (>23kb and 4.3 kb).CONCLUSION The rearrangement of VEGF in the ovary cancer overexpressing VEGF does exist.
