大肠癌中microRNA-93的表达及其与临床病理因素的关系

摘　　要：目的探讨大肠癌中microRNA-93的表达及其与临床病理因素的关系。方法取42例大肠癌及癌旁正常黏膜组织标本,采用茎环逆转录实时荧光定量PCR方法检测大肠癌及癌旁正常黏膜组织中microRMA-93的表达水平,并比较分析其在不同临床病理因素间的表达差异。结果(1)microRNA-93在大肠癌组织中的表达水平显著高于癌旁正常黏膜组织,差异有统计学意义(P<0.01);(2)microRNA-93的表达与大肠癌淋巴结转移和浸润深度有关(P<0.05或0.01);而与患者的年龄、性别、肿瘤大小、远处转移、分化程度无关(均P>0.05);(3)microRNA-93评价大肠癌淋巴结转移的敏感性和特异性分别为0.867和0.556,评价大肠癌浸润深度的敏感性和特异性为0.800和0.833。结论 microRNA-93表达上调可能与大肠癌的发生、发展有关,microRNA-93可作为评价大肠癌淋巴结转移和浸润深度的辅助指标。Objective To investigate the expression of microRNA-93 in colorectal carcinoma and its relation to clinico- pathological features of the cancer. Methods The expression of microRNA-93 stem-loop real-time RT-PCR in cancer tissue and matched pericancerous tissue specimens from 42 patients with colorectal carcinoma. The relationship between microR- NA-93 expression and clinicopathologic features of colorectal carcinoma was analyzed. Results The expression level of mi- croRNA-93 was significantly higher in colorectal carcinoma than that in pericancerous tissues （P〈0.05）. The expression of mi- croRNA-93 was positively correlated with the lymph node metastasis and depth of tumor invasion （P〈0.05 or 0.01）, not with age, sex, tumor size, distant metastasis and cell differentiation（P 〉0.05）. The sensitivity and specificity of microRNA-93 expression for evaluation of lymph node metastasis were 0.867 and 0.556 respectively; those for evaluation of tumor invasion depth were 0.800 and 0.833, respectively. Conclusion The over-expression of microRNA-93 may be associated with development and progres- sion of colorectal carcinoma. The expression of microRNA-93 might be used as a adjuvant parameter for predicting the lymph node metastasis and depth of tumor invasion in colorectal carcinoma.
