摘  要:目的 :人工角膜支架材料植入角膜板层囊带后 ,观测GAGs在角膜及支架材料中的沉积及合成。方法 :将人工角膜支架材料制成圆盘状以手术方法植入角膜板层囊带内 ,对侧未手术眼及仅制备囊带眼分别为正常对照及手术对照。GAGs经 4M guanidin HCI提取 ,提取物经Q sepharose柱层析。应用选择性多糖酶消化测定GAGs。结果 :人工角膜支架材料植入角膜实质层后 ,角膜总GAG的含量降低 ,而DS CS/KS ,HS/KS比率在周围组织和材料中增加。结论 :将人工角膜支架材料植入角膜实质层 ,引起材料内和周围角膜组织GAGs的组成及硫酸化的改变。术后 84d材料内GAGs的组成与周围角膜组织相似 ,表明支架材料逐渐被周围角膜组织所接受。Objective:To examine the synthesis and deposition of corneal glycosaminoglycan (GAGs) in response to a wound created by the insertion of porous disc into stromal lamealar pockets.Methods:The disc and surrounding stromal tissue were assayed and compared to corneas at 14,42 and 84 days.GAGs were extracted with 4M guanidine HCI and chromatographed on Q sepharose columns.Bound proteoglycans were eluted with a linear salt gradient.Total GAG were determined by enzyme digeotion with selective polysaccharide lyases.Results:The placement of the discs into the stroma resulted in a decrease in total amount of GAG.However,the ratio of dermatan chondroitin sulfate and heparan sulfate to keratan sulfate increased in the surrounding tissue and disc.Conclusion:The insertion of discs into intetamellar pockets causes changes in the sulfuration and proportion of the glycosaminoglycans in the surrounding tissue and disc.84 days later,the population of glycosaminoglycans in the disc begin to resemble the surrounding stroma.This means the discs are accepted by cornal stromal tissue.
