摘  要:该文研究了奥硝唑羧甲基壳聚糖纤维膜植入小鼠体内的组织相容性,以探讨其在牙周炎辅助治疗中的安全可靠性。首先利用静电纺丝技术将载有奥硝唑微球的羧甲基壳聚糖"串珠"丝状纤维制备成膜,再用此膜进行小鼠动物实验。选择羧甲基壳聚糖膜和医用水溶性淀粉膜为对照组,将3种膜剪为方形分别埋入3组小鼠股部肌肉,观察并记录各组小鼠埋入部位和全身反应,并在第7d、14d、21d分别取埋入局部肌肉组织,经2.5%戊二醛固定,用常规透射电镜制片、光镜和电镜观察小鼠肌肉组织及其主要脏器的组织超微结构改变。结果表明:羧甲基壳聚糖膜对组织刺激有轻微炎症反应,但较水溶性淀粉膜更温和、更安全,奥硝唑羧甲基壳聚糖膜的刺激反应介于两者之间。羧甲基壳聚糖具有良好组织相容性,该文为奥硝唑羧甲基壳聚糖纤维膜应用于口腔临床提供了参考。The safety and reliability was investigated when ornidazole carboxymethyl ehitosan fihers were applied to adiuvant treatment of periodontitis through studying the stocompatibility when the fibers were implanted in mice. Electrospinning technique was used to prepare microspheres containing ornidazole with three kinds of membranes prepared for experiments, carboxymethyl chitosan fiber membranes with "beaded" filamentous structures, single carboxymethyl chitosan films, and treated water-soluble starch films. These three kinds of trimmed square-shaped films were embedded into thigh muscles of three groups of mice with the embedded parts and total physical responses recorded. The localmuscle tissues near the embedded parts were then taken on the 7th day, the 14th day, and the 21st day. After fixed with 2.50 glutaraldehyde, the muscle tissue of the mice, the ultrastructure of main organs and tissue changes were observed using transmission electron microscopic technology. The results show that carboxymethyl chitosan membrane tissue stimulation causes slight inflammation which is more moderate and more secure than that of the starch films, while the stimulus response of ornidazole carboxymethyl chitosan membranes is between them. Preliminary evaluation indicates that carboxymethyl chitosan has good biocompatibility, which shows that ornidazole carboxymethyl chitosan fiber membranes provide a promising choice for oral clinical application.
