利用细胞模型和化学法评价核桃叶黄酮的抗氧化活性

摘  要:从八种大孔树脂中筛选出了适宜纯化核桃叶黄酮的大孔树脂,并对纯化后核桃叶黄酮的细胞内抗氧化活性、对羟自由基清除力、对DPPH自由基清除力和对超氧阴离子自由基清除力进行了研究。结果表明,AB-8型大孔树脂的纯化效果最佳,核桃叶黄酮具有明显的抗氧化活性。核桃叶黄酮对羟自由基清除率高于VC,对超氧阴离子自由基抑制率和DPPH自由基清除率低于Vc,其IC50为22.4μg/mL。核桃叶黄酮在结肠癌细胞(HT-29)中表现出明显的抗氧化活性,其抗氧化性随浓度递增。The adsorption and desorption of eight macroporous resins for flavonoids from Juglans regia leaves were compared.The optimal adsorbent was AB-8.Moreover,the ability of purified flavonoidson cellular antioxidant,scavenging hydroxyl radical (· OH),scavenging 1-diphenyl-2-picrylhydrazy (DPPH) and superoxide anion radical (62-) was investigated.The results showed that flavonoids have good antioxidant activity.Compared with VC,the flavonoids from Juglans regia leaves had stronger capacity of scavenging on OH and 62-,but weaker capacity of scavenging on DPPH with IC5o 0.0224mg/mL.Furthermore,the flavonoids showed obvious antioxidant activity in colon cancer cell (HT-29),and it was concentration-dependent.
