重庆地区汉族儿童β_(2)-肾上腺素能受体单倍型与哮喘相关性的研究

摘  要:目的 探讨 β2 肾上腺素能受体 (β2 AR)单倍型与重庆汉族哮喘儿童哮喘的相关性。方法 采用PCR 限制性酶切分析法检测 76例汉族哮喘儿童 β2 AR16、2 7位多态性 ,并与 10 0例健康人进行对照 ,统计分析 β2 AR单倍型与哮喘病情严重度的关系。结果 对照组 β2 AR甘氨酸 (Gly) 16及谷氨酰胺 (Gln) 2 7等位基因频率分别为 0 .42、0 .70 ,哮喘组为 0 .5 0、0 .72 ,后者与对照组比较 ,无显著差异 (P >0 .0 5 )。Gly16等位基因及Gly16 Gln2 7单倍型在重度哮喘组中频率分别为 0 .71、0 .47,在 3组中的分布差异显著 (P<0 .0 1,P <0 .0 5 ) ,以重度哮喘组分布频率较高。结论 Gly16及Gln2 7型 β2 AR多态性不是中国重庆汉族儿童哮喘发病的主要因素 ,但Gly16等位基因与Gly16 Gln2Objective To study the correlation between β 2 adrenergic receptors (β 2 AR) haplotypes and asthma in the children of the Han nationality in Chongqing. Methods A total of 76 unrelated asthmatic children and 100 healthy individuals (50 children and 50 adults) were subjected from the Han nationality of Chongqing. The polymorphisms of locus 16 and 27 of the β 2 AR from all subjects were studied with PCR restriction fragment analysis. Statistical analysis was carried out on the correlation among glycine (Gly) 16 allele, Gly16/glutamine (Gln) 27 haplotype and the degree of severity of the clinical status of asthma. Results The distribution frequency of Gly 16 and Gln 27 allele accounted for 0.42 and 0.70 in the control group, 0.50 and 0.72 in asthmatic group respectively. There were no significant increase of the frequency of Gly16 and Gln27 allele in the asthmatic group compared with control group. The distribution frequency of Gly16 allele and Gly16/Gln27 haplotype in severe asthmatic group accounted for 0.71 and 0.47 respectively. Significant increase was found in the frequency of Gly16 allele and Gly16/Gln27 haplotype in the severe asthmatic group than the mild and moderate asthmatic groups ( P <0.01, P <0.05 respectively). Conclusion Asthma is not primarily caused by the polymorphisms of Gly and Gln alleles of β 2 AR, but Gly16 allele and Gly16/Gln27 haplotype are possibly correlated with asthmatic severity in the children of the Han nationality in Chongqing.
