摘　　要：目的通过研究肺癌患者组织中甲状腺转录因子-1(TTF-1)是否存在异常高表达,进而探讨其表达在不同肺癌类型之间的联系,为实现肺癌的生物学疗法提供理论依据。方法收集2009年1月至2010年12月本院经手术切除及纤维支气管镜镜下活检的43例肺癌组织及外周血标本,使用免疫组织化学S-P法研究其组织中TTF-1的表达,在表达阳性且经统计学检测TTF-1在肺癌组织者表达异常的情况下,应用RT-PCR方法进一步求证患者外周血单个核细胞中TTF-1的基因表达。结果 TTF-1在肺癌组织及外周血单个核细胞中的表达明显异常,在不同病理类型癌组织中的表达存在明显差异(χ2=9.242,P=0.026<0.05),且与肺癌分化程度有关(χ2=15.172,P=0.01<0.05)。结论肺癌患者癌组织TTF-1表达与肺癌有无淋巴结转移和分化程度关系密切,随病变进展及分化程度的降低而增高,可作为预测肺癌转移及预后评估的指标。Objective This study was aimed to evaluate the prevalence and prognostic implication of thyroid transcrip- tion factor-1 （ TTF-1 ） immunoreactivity in 43 human lung carcinomas, and to investigate its linkages between the differ- ent lung cancer types. Method From January 2009 to December 2010, after surgical resection and bronchoscopy biopsy 40 cases of lung cancer tissue and peripheral blood specimens. SP immunohistochemical method to study the expres- sion of TTF-1 in their organization, expression in ling cancer organizer application in exceptional circumstances （RT- PCR） method to further verify the statistical detection of TIT-1 expression in lung cancer organizer applications in ex- ceptional circumstances（RT-PCR） method to further verify the peripheral blood of patients with TIT-1 gene expres- sion. Result The lung cancer tissue with the degree of differentiation of lung cancer. Conclusion： the lung cancer tis- sue ,TFF-1 expression in lung tissue and blood are obvious abnormality, and there are significant differences in expres- sion in lung cancer. Conclusion The lung cancer tissue,TTF-1 expression in lung cancer lymph node metastasis and closely related to the degree of differentiation, increased with disease progression and reduce the degree of differentia- tion,can be used as a predictor of lung cancer metastasis and prognosis indicatiors.
