摘  要:目的:研究紫草素(Shikonin)对Burkitt's淋巴瘤细胞凋亡及坏死的影响及Necrostatin-1(Nec-1)对紫草素诱导的细胞凋亡的作用。方法:对体外培养的Raji细胞进行荧光显微镜观察,Alarma Blue法检测细胞增殖率,流式细胞仪分析细胞凋亡及坏死,免疫印迹检测相关蛋白表达。结果:紫草素对Raji细胞生长有抑制作用,呈时间和浓度依赖。紫草素可诱导Raji细胞发生程序性坏死及凋亡。小分子化合物Nec-1不仅可以抑制紫草素诱导的程序性坏死,同时对细胞凋亡也有抑制作用。结论:紫草素可以抑制Raji细胞生长增殖,而小分子Nec-1可以抑制这一过程。Objective:To explore the role of Shikonin in Burkitt's lymphoma cell death and the effect of Necrosta- tin - 1 (Nec - 1 ) in this process. Methods: Shikonin and Nec - 1 were used to induce or inhibit cell death of Raji B cell. The proliferation inhibition rate, apoptosis and necrosis rate, and death related protein expression of Raji B cells were measured by Alarma Blue assay,flow cytomett7 and Western blot respectively. Results:Shikonin inhibited the growth of Raji cell in a time - and concentration - dependent manner and contributed to apoptosis and necrosis of Rail cell. Nec - 1 inhibited both the apoptosis and necroptosis of Raji B cell caused by Shikonin. Conclusion:Shikonin causes Raji cell death which is inhibited by Nec - 1.
