鸡传染性鼻炎PCR和阻断ELISA试剂盒的研制与应用

摘　　要：用鸡传染性鼻炎 PCR试剂盒可直接检测病料中的病原 DNA，可测出 1 pg的 DNA和 102～ 103个细菌，对可疑病料的检出率是传统的细菌分离方法的 2～ 3倍，可重复性好，特异性为 100％，在 4℃可保存 12个月，在－ 20℃可保存 15个月。阻断 ELISA试剂盒可检测副鸡嗜血杆菌型特异性抗体，其种特异性为 100％，重复性好。试剂盒在 37℃可保存 7 d以上， 4℃可保存 10个月，－ 20℃可保存 1年以上。用两个诊断试剂盒检验 155份可疑病料、 2 191份血清， PCR的阳性率为 32.3％； A型抗体阳性率为 27.4％， C型抗体阳性率为 11.5％。According to PCR and Blocking ELISA(B－ ELISA)methods for infection coryza(IC) established by our laboratory,we developed both PCR and B－ ELISA kits for IC.PCR kit can detect 1 pg DNA of the clinical pathogen directly with the minimal content of 1 pg DNA and 102～ 103 cells.Its specificity is complete.It kept effective until stired at－ 20℃ for 15 months at least and at 4℃ for 12 months.Detecting positive with the kit is 2～ 3 higher times than that with traditional bactieria culture.B－ ELISA kit has a complete specificity and better sensitivity.It kept effective until stored at－ 20℃ for 12 months at least and at 4℃ for 10 months. In Beijing、 Shandong、 Hebei and other places we collected 155 clinical samples and 2 191 sera samples 32.3％ of the clinical samples were positive with PCR kit, 27.4％ of the clinical sera samples were positive for type A and 11.5％ for type C with B－ ELISA kit.
