摘 要:聚磷菌在强化生物除磷中起关键作用,与聚磷菌新陈代谢密切相关的胞内聚合物有PHAs、糖原和poly-P。聚磷菌胞内聚合物的定性和定量测定方法有很多。文章采用多种方法对多聚物PHAs和poly-P进行染色,结果显示,对厌氧阶段生成的PHB颗粒的染色,尼罗蓝染色法比苏丹黑染色法特异性高;好氧阶段生成的多聚磷酸盐的染色方法中,DAPI染色法和奈瑟氏新方法效果更好。Polyphosphate-accumulating microorganisms (PAOs) plays key roles in enhanced biological phosphorus (P) removal. Intracellular polymers, including polyphosphate (poly-P), polyhydroxyalkanoates (PHAs) and glycogen, were proved to have close relations with PAO's metabolism. PAOs storage polymers can be identified and quantified by many methods. In this paper, a variety of methods were used to dye PAOs' storage polymers. Results showed that Nile Blue staining was more specific than Sudan black staining to the PHB in anaerobic stage, and to the poly-P in aerobic, DAPI staining and Neisser new staining were proper than others.
