摘 要:目的观察羟基喜树碱(hydroxycamptothecin, HCPT)单独及与甲氧胺(methoxyamine,MX)联合用药在抑制神经胶质瘤(SHG44)胶质瘤细胞增殖过程中是否具有协同作用,以期减少HCPT用量,降低毒副反应,为HCPT联合MX应用于抗胶质瘤治疗提供理论支持。方法联合运用HCPT与碱基切除修复抑制剂MX,用MTT法、流式细胞等方法检测联合用药的体外抑瘤效果及可能的机制。结果 HCPT联合甲氧胺能显著抑制SHG44胶质瘤细胞增殖,随着浓度增加和作用时间延长,其增殖抑制效应也随之增强。流式细胞检测结果表明,HCPT联合MX组凋亡细胞比例明显增多,但与单独HCPT用药组比较未见明显差异。结论 HCPT联合MX对SHG44胶质瘤细胞具有显著增殖抑制作用,且具有明显剂量-时间依赖性。Objective To improve the glioma suppression effect of hydroxycamptothecin (HCPT)and reduce its side-effects by changing its pharmaceutical delivery form. To clarify whether a synergetic effect exists between HCPT and methoxyamine (MX) on the inhibition of glioma cell proliferation and provide theoretical basis for the further usage of HCPT in the treatment of glioma. Methods MTT and flow cytometry (FCM) were applied to determine the growth inhibition effect of using HCPT alone or in combination with MX on SHG4 cell line. Results Different concentrations of HCPT and MX were used alone or in combination and this combination usage showed a higer inhibition rate than using HCPT or MX alone. FCM results showed more cells were stopped at the G2 phase in the HCPT+ MX group. However, the difference was not statistically significant between HCPT+ MX group and HCPT group. Conclusion HCPT could inhibit the proliferation of SHG44 cell and the combination of MX could increase this inhibitory effect on SHG44, suggesting a synergistic effect between HCPT and MX.
