摘　　要：目的:观察不同浓度转化生长因子β1(TGF-β1)对绒毛膜癌JEG-3细胞侵袭能力的影响及尿激酶型纤溶酶原激活物(uPA)及其特异性受体(uPA receptor,uPAR)和纤溶酶原激活物抑制剂1(PAI-1)的蛋白表达,进一步探讨TGF-β1在绒毛膜癌发生发展中的作用机制。方法:分别取不同浓度的TGF-β1作用于绒毛膜癌JEG-3细胞,采用Transwell侵袭实验检测细胞的侵袭能力变化,蛋白印迹法检测uPA、uPAR、PAI-1蛋白的表达差异。结果:①0μg/L、1μg/L、10μg/L和100μg/L TGF-β1作用于绒毛膜癌JEG-3细胞后,各组细胞的穿膜细胞数明显增多(P<0.05),且具有浓度效应;②0μg/L、10μg/L、50μg/L、100μg/L和200μg/L TGF-β1作用于绒毛膜癌JEG-3细胞后,各组细胞中uPA、uPAR、PAI-1的蛋白表达明显升高(P<0.05),且具有浓度效应。结论:外源性TGF-β1促进了绒毛膜癌JEG-3细胞的侵袭能力,增强uPA、uPAR、PAI-1蛋白表达的受诱导程度,且表现出良好的浓度依赖效应。Objective： To observe the invasive ability of choriocarcinoma JEG -3 cells and the effects on protein expressions of uPA, uPAR, PAl - 1, caused by different concentrations of transforming growth factor -β1 （TGF - β1 ） . Methods： Transwell invasive experiment was used to dectect the invasive ability, and Western blot was used to dectect uPA, uPAR and PAI - 1 protein expressions in Choriocareinoma JEG - 3 cells induced by TGF - β1 with different concentrations. Results： （1）The amount of transmigrated cells of Choriocarcinoma JEG -3 was increased directly after TGF - β1 induction with 0, 1, 10 and 100μg/L respectively （P 〈0. 05）, with a significant concentration - dependent effect. （2）The protein expressions of uPA, uPAR and PAl - 1 were obviously increased by treating with different concentrations of TGF - β1 at 0, 10, 50, 100 and 200 μg/L （P 〈 0. 05）, with concentration - dependent manner. Conclusion： External source TGF - β1 can promote the invasive ability of Choriocarcinoma JEG - 3 cells and increase the induction level of protein expression of uPA, uPAR, PAI - 1. It presents a good concentration - dependent effect.
