摘　　要：目的：观察在人绒癌JEG-3细胞株中应用短发夹RNA抑制存活素表达的效果。方法：选取已证实有效的靶向Survivin的shRNA序列，将其转染JEG-3细胞后检测SurvivinmRNA水平的变化，同时进行荧光报告检查。结果：shRNA表达载体构建成功，转染JEG-3报告细胞后转染组24h孔中均有弱荧光表达，48h荧光表达增强，转染率60％-75％，测序证实构建的表达载体序列正确，转染后转染组JEG-3细胞中的Survivin mRNA和蛋白的表达明显减弱。结论：靶向Survivin短发夹RNA表达载体构建成功，并能有效下调Survivin的表达水平。Objective： To observe the expression of survivin in human choriocarcinoma JEG -3 cell lines and the expression of surviving inhibited by short hairpin RNA . Methods： Proven effective targeting Survivin shRNA sequence was selected, transfected into JEG -3 cells. Survivin mRNA level changes were detected and fluorescent reporter was checked. Results： shRNA expression vector was suc- cessfully constructed and transfected into JEG -3 cells and there was weaker fluorescence intensity. Sequencing confirmed that the constructed expression vector sequence was correct. The expression of Survivin was inhibited at the mRNA level. Conclusion： Targeting Survivin small interfering RNA expression vector is successfully constructed and can effectively down - regulat the expression of Survivin gene.
