摘 要:目的 观察促凋亡基因程序性细胞死亡5基因(PDCD5)在胆道闭锁(BA)及胆总管囊肿(CC)患儿胆管组织中的表达,探讨促凋亡基因PDCD5在BA发病机制中的作用.方法 分别取18例BA和11例CC患儿的胆管组织,应用Westem blot与荧光实时定量聚合酶链反应(FQPCR)方法分别检测PDCD5在BA与CC患儿胆管组织中的表达,对其表达进行定位和定量分析.结果 PDCD5在CC组胆管组织中的Ct值为(6.7292±1.1690),高于BA组的相应CT值(4.012 5±1.073 5,P<0.05).应用2-△△CT计算出CC与BA中PDCD5基因表达量的比值为1.00∶6.57.PDCD5在BA组胆管组织中mRNA转录水平明显高于对照组;Western blot结果显示,PDCD5在BA组胆管组织中蛋白表达量明显高于CC组.结论 PDCD5在BA患儿胆管组织中的蛋白和基因水平表达均增强.PDCD5可能参与BA的胆管上皮细胞的凋亡,从而参与BA的炎性反应.Objective To characterize the expression and significance of programmed cell death 5 (PDCD5) gen in bile duct tissues of biliary atresia (BA) and choledochal cyst (CC).Methods Small pieces of bile duct tissues were obstained from 18 patients with BA and 15 patients with CC.The expression of PDCD5 protein and mRNA was detected by using Western blotting and real-time fluorescent quantitative polymerase chain reaction(FQ-PCR) respectively.Results In the BA patients,the CT of PDCD5 in bile duct tissues was (4.012 5 ± 1.073 5),significantly lower than in the CC patients (6.729 2 ± 1.169 0)(P〈0.05).The ratio of PDCD5 in CC and BA is 1.00∶6.57 by 2-△△CT.Above all,the expression of PDCD5 mRNA was enhanced in bile duct tissues of BA.The expression of PDCD5 protein in the BA patients was higher than in the CC patients.Conclusion The expression of PDCD5 protein and mRNA was enhanced in bile duct tissues of BA,suggesting that PDCD5 may be involved in the apoptosis of bile duct epithelia cells in BA and participate in BA inflammatory reaction,which plays an important role in the pathogenesis of BA.
