摘  要:目的对依达拉奉清除自由基的效果做即时可视化评价。方法通过使用·O2-荧光探针(MitoSOXTM)或·OH荧光探针(HPF)标记大鼠脑表区域神经细胞,共焦点激光显微镜活体观察在大鼠缺血(10min)再灌注(30min)过程中,脑表区域·O2-或·OH产生的情况,比较模型对照组与依达拉奉治疗组(3.0mg/kg缺血前静脉给药)之间·O2-或·OH产生情况的差异,对依达拉奉的自由基(·O2-,·OH)清除效果进行可视化评价。结果显示缺血开始3min后,·O2-及·OH产生开始,并一直持续增加到再灌注后10min达到峰值;依达拉奉有效清除·OH产生,而对·O2-的清除效果不明显。结论依达拉奉在缺血期及再灌注早期能有效地清除·OH的生成。Objective To investigate the effect of edaravone on the reactive oxygen species elimination by a real-time intravital fluorescence imaging .Methods In anesthetized adult rats ,a fluorescent dye ,MitoSOXTM for superoxide radical (·O2-),and hydroxyphenyl fluorescein for hydroxyl radical (· OH) was injected into cortices by a pressurized bolus .Through a closed cranial window ,fluorescent images were taken with a confocal micro-scope on 10 minutes forebrain ischemia and 30 minutes reperfusion .We also investigated the effect of edaravone on the reactive oxygen species elimination by this intravital fluorescence imaging .Results In ischemia and the early period of reperfusion ,the fluorescence intensity significantly increased (to 183% in MitoSOX and to 189% in hydroxyphenyl fluorescein) ,and edaravone (3 .0 mg/kg) treatment completely scavenged · OH ,but did not do so in · O2-generation .Conclusion ROS production increased in ischemia and the early period of reperfusion and edaravone can selectively eliminate the · OH .
