摘 要:目的探讨电针刺激和骨髓间充质干细胞(BMSC)移植联合治疗对脑出血大鼠出血灶周边区基质金属蛋白酶9(MMP-9)及血清白细胞介素2(IL02)表达的影响。方法向成年SD大鼠右侧尾壳核注射肝素和胶原酶诱导脑出血模型,并于术后第3天向出血灶移植BMSC后施以电针刺激(Ea.BMSC组),同时设置BMSC移植组(BMSC组)、电针刺激组(Ea组)和生理盐水注射组(NS组)作为对照。各组分别于治疗后1、3、5、7和14d收集血清和脑组织标本,采用免疫荧光法检测出血灶周边区MMP-9的表达变化,放射免疫法检测血清IL-2的含量。采用单因素方差分析中的LSD法进行统计分析。结果Ea—BMSC组的MMP-9表达在1~7d时低于NS组(t值分别为9.85、5.76、7.05、3.13,P均〈0.05),在全部时间点均低于BMSC组(t值分别为8.91、11.68、11.20、10.11、2.76,P均〈0.05),而在3—14d时则高于Ea组(t值分别为-3.38、-3.98、-5.66、-13.07,P均〈0.05)。Ea.BMSC组的血清IL-2含量在5d和7d时高于Ns组(t值分别为-4.15、-3.81,P均〈0.05),在3d和5d时分别低于和高于BMSC组(t值分别为2.76、-3.69,P均〈0.05),在3d和14d时分别高于和低于Ea组(t值分别为-2.90、5.64,P均〈0.05)。结论电针联合BMSC移植治疗可抑制因单纯BMSC移植治疗所引起的MMP-9表达增高,且较电针组和BMSC组更能使IL-2含量保持在较稳定的水平。Objective To explore the effects of electro-acupuncture(Ea) stimulation combined with bone marrow mesenehymal stem cells (BMSC)transplantation on the expression of matrix metalloproteinase-9 ( MMP-9 ) and interleukin-2 ( IL-2 ) in rats after intracerebral hemorrhage ( ICH ). Methods ICH rats were replicated by injecting the collagenase and heparin into right caudate putamen. After 3 days, the rats were treated by BMSC transplantation and electro-acupuncture stimulation (Ea-BMSC group), or dealed with BMSC transplantation( BMSC group), electro-acupuncture stimulation (Ea group)and physiological saline injection ( NS group) respectively. The serum and brain tissues of rats were collected at 1,3,5,7 and 14 d after treatment. Immunofluorescence and radioimmunoassay were used to detected expression of MMP-9 and IL-2 respectively. LSD method was performed for statistical analysis. Results Expression of MMP-9 in Ea-BMSC group was lower than that in NS group at 1-7 d(t =9.85,5.76,7.05,3.13,all P 〈0. 05) ,and lower than that in BMSC group at all time points(t = 8.91,11.68,11.20,10. 11,2.76,all P 〈0. 05) ,but higher than that of Ea group .at 3-14 d ( t = - 3.38, - 3.98, - 5.66, - 13.07, all P 〈 0. 05 ). The content of IL-2 inserum in Ea-BMSC group was higher than that of NS group at 5 d and 7 d(t = -4. 15, -3.81,all P 〈 0. 05 ), and lower and higher than that in BMSC group at 3 d and 5 d respectively ( t = 2. 76, - 3.69, all P 〈 0. 05) ,and higher and lower than that in Ea group at 3 d and 14 d respectively(t = -2.90,5.64,all P 〈 0. 05). Conclusions Electro-acupuncture combined with BMSC transplantation can inhibit MMP-9 increase caused by simple BMSC transplantation, and helps to maintain the content of IL-2 at a relatively stable level compared to Ea group and BMSC group.
