摘  要:目的建立检测患者全血中环孢素A浓度的高效液相色谱法。方法血样经乙醚提取后,以C18色谱柱为固定相,乙腈-甲醇-水(72:3:25)为流动相,60℃柱温下进行色谱分析,紫外检测波长为210nm。结果CsA全血浓度在50~1500ng/mL范围内呈良好的线性关系,检测限为25ng/mL,批内和批间精密度RSD均小于10%,方法的绝对回收率为60.14%~69.04%。结论HPLC法快速准确,与FPIA法相关性良好,均可用于肾移植术后患者CsA的血药浓度监测。Objective To develop an HPLC method for determining the concentration of cyclosporine A (CsA) in whole blood of renal transplant patients. Method Detection was performed on C ls column with acetonitrile: methanol: water (72: 3:25 ) as mobile phase. Detecting wavelength was 210 nm, and column temperature was 60 ℃. Results The calibration curve was linear in the content rations ranged from 50 - 1 500 ng/mL. Absolute recovery of CsA was 60.14% -69.04%, the inter-day and the intra-day RSD was less than 10% (n = 5 ) . Conclusion This IqPLC method is simple and reliable, as the same as FPIA method. It can be applied to determine the concentration of CsA in whole blood of patients receiving transplantation.
