Fas/FasL基因多态性与乳腺癌发生的相关性

摘　　要：目的：探讨Fas/FasL基因多态性与汉族人群乳腺癌发病风险的关联性。 方法：提取250例乳腺癌患者和250例体检健康者外周血基因组DNA，用聚合酶链反应-限制性片段长度多态性（PCR-RFLP）分析技术对Fas基因1377G〉A,670A〉G和FasL基因844T〉C位点多态性进行基因分型，用非条件Logistic回归方法计算风险比（OR）及95%可信区间（95%CI）。 结果：与Fas 1377多态性位点GG基因型相比，GA、AA基因型显著降低乳腺癌的发病风险［校正OR（95%CI）分别为0.44（0.29~0.68），0.29（0.17~0.48），P均〈0.01］。与Fas 670多态性位点AA基因型相比，AG、GG基因型显著降低乳腺癌的发病风险［校正OR（95%CI）分别为0.63（0.42~0.94）和0.36（0.21~0.60），P均〈0.01］。与FasL 844多态性位点CC基因型相比，TC基因型显著降低乳腺癌的发病风险［校正OR（95%CI）为0.58（0.40~0.84），P=0.012）。 结论：Fas-FasL基因多态性与乳腺癌发病风险相关。Objective：To investigate the correlation of Fas and FasL genes polymorphism with the breast cancer risk in Han population. Methods：Genomic DNA was extracted from the peripheral blood mononuclear cells （PBMCs） of 250 patients with breast cancer and 250 healthy controls. Then, the genotypes of Fas 1377G〉A, 670A〉G and FasL 844T〉C were determined by the PCR-RFLP method and Logistic regression analysis was used to calculate the odds ratio （OR） and 95% confidence interval （95%CI）. Results Compared with the Fas 1377 GG genotype, GA and AA genotypes obviously decreased the breast cancer risk （adjusted OR=0.44, 95%CI： 0.29 0.68 for GA; adjusted OR=0.29, 95%CI： 0.17-0.48 for AA, P〈0.01）. Similarly, compared with the Fas 670AA genotype, AG and GG genotypes also significantly decreased the breast cancer risk （adjusted OR=0.63, 95%CI： 0.42-0.94 for AG; adjusted OR=0.36, 95%CI： 0.21-0.60 for GG, P〈0.01）. In addition, compared with the FasL 844CC genotype, TC genotype significantly decreased the breast cancer risk （adjusted OR=0.58, 95%CI： 0.40 0.84, P=0.012）. Conclusion：The polymorphisms of Fas and FasL genes are correlated with the breast cancer risk.
