摘  要:目的研究放线菌SIIA243对维生素D3的羟基化作用。方法将VD3加入到不同菌株的培养液中,通过高效液相色谱检测培养液中VD3的转化产物以确定目标菌株,然后在50升发酵罐中培养目标菌株,添加维生素D3,发酵完成后对转化产物进行分离纯化和结构鉴定。结果发现菌株SIIA243转化产物的高效液相图谱中,转化产物1的保留时间与25OHVD3基本相同,转化产物2的保留时间与1α,25(OH)2VD3基本相同;经紫外光谱、质谱和核磁共振谱分析,纯化后得到的两个转化产物分别鉴定为25OHVD3和1α,25(OH)2VD3。结论菌株SIIA243能够将维生素D3转化为25OHVD3和1α,25(OH)2VD3。Objective Study on hydroxylation of vitamin D3 by an actinomycetes SIIA243. Methods Vitamin D3 was incubated with growing culture of different strains, the transformation products from vitamin D3 were detected by HPLC in order to find the target strain.Then, the target strain was cultured in a 50L fermentor and vitamin D3 was added. After completion of fermention, the transformation products were purled and the structures were elucidated. Results According to the HPLC chromatogram of transformation products of strain SIIA243, the retention time of the transformation product 1 was approximately the same as the retention time of 25OHVD3, the retention time of the transformation product 2 was approximately the same as the retention time of 1α, 25(OH)EVD3, two purified transformation products were separately elucidated as 25OHVD3 and 1α, 25(OH)EVD3 by means of spectral analysis including UV, MS and NMR. Conclusion Strain SIIA243 was able to convert vitamin D3 to 25OHVD3 and 1α, 25(OH)2D3.
