摘 要:目的 研究胃癌组织中脆性组氨酸三位体 (fragilehistidinetriad ,FHIT)基因转录的改变。方法 采用逆转录 聚合酶链反应 (RT PCR)方法对 43例胃癌组织及其相应癌旁非癌组织进行FHIT基因cDNA检测。结果 除5例胃癌、2例非癌组织缺失FHIT基因野生型cDNA外 ,其余标本均扩增出FHIT基因野生型cDNA片段。 2 2例(5 1.2 % )胃癌、9例 (2 0 .9% )的癌旁非癌组织扩增出FHIT基因异常cDNA片段 ,二者统计学差异有显著性 (P <0 .0 5 )。Objective To investigate alterations of fragile histidine triad (FHIT) gene in human gastric cancer. Methods Tumor tissues and adjacent tissues in 43 cases of gastric cancer were examined for abnormalities of the gene by using the reverse transcriptase polymerase chain reaction (RT PCR). Results Wild type FHIT gene transcripts were found in all samples except 5 tumor tissues and 2 adjacent tissues. Abnormal FHIT gene transcripts were obtained from 51.2% (22/43) of gastric cancers and 20.9% ( 9/43 ) of their nearby tissues. The rate of aberrant FHIT transcript in cancerous tissues was significantly higher ( P <0.05 ) than that in their adjacent tissues. Conclusion The aberrant transcripts of FHIT gene maybe related with the development of gastric cancer.
