摘 要:目的检测转录因子维甲酸相关孤核受体α(retinoid acid receptor related orphan receptorα,RORα)、细胞表面受体T1/ST2和IL-17RB以及IL-5和IL-13等细胞因子的表达水平;了解胃癌患者外周血中nuocytes细胞的极化状态,探讨其与胃癌发生发展的关系。方法常规Ficoll-泛影葡胺密度梯度离心法分离外周血单个核细胞;实时荧光定量PCR检测30例胃癌患者PBMC中RORα、IL-17RB、T1/ST2、IL-5、IL-13 mRNA表达水平。结果胃癌患者外周血PBMC中RORα、IL-17RB、T1/ST2、IL-5 mRNA表达水平明显高于健康对照组,而IL-13的表达水平则明显低于健康对照组。相关性分析结果表明,胃癌患者RORαmRNA水平与IL-17RB、T1/ST2、IL-5、IL-13 mRNA水平呈正相关。结论胃癌患者机体存在着nuocytes极化状态,可能是构成胃癌患者Th1/Th2细胞平衡失调的基础,也与MDSC、M2巨噬细胞参与的免疫抑制微环境有着密切的关系。This study designed to clarify" the nuocytes polarization state and explore its relationship with the development of gastric cancer (GC), through detecting the expression levels of transcription factor RORα, receptors IL-17RB and T1/ST2, and cytokines IL-5 and IL-13 in peripheral blood from patients with gastric cancer. Quantitative real-time PCR (qRT-PCR) was used to determine the mRNA expression levels of RORα, IL-17RB, T1/ST2, IL-5 and IL-13 in peripheral blood mononuclear cell (PBMC). Our data showed that the expression levels of RORα, IL-17RB, T1/ST2 and IL-5 mRNA in PBMC from GC patients were higher as compared with healthy controls, while the IL-13 mRNA expression levels was significantly decreased. In addition, there was a positive correlation between RORα and IL-17RB, T1/ST2, IL-5 or IL-13. All the result indicated that the existence of nuocytes polarization in the patients with gastric cancer may contribute to the Thl/Th2 cell imbalance and closely relate to suppressive microenvironment involving MDSC and M2 macrophage.
