摘  要:目的通过比较不同处理纯钛表面对人牙龈成纤维细胞粘附及增殖的影响,探讨更有利于其附着的种植体颈部表面处理方法。方法制备直径10mm、厚度1mm,Ra 0.8μm的纯钛片,按表面处理方法分为三组,A组:TiO2喷砂双重酸蚀组,B组:Al2O3喷砂双重酸蚀组,C组:光滑组。将体外培养的人牙龈成纤维细胞分别接种于各组钛片表面,扫描电镜观察人牙龈成纤维细胞形态与分布,MTT法测定各组钛片表面人牙龈成纤维细胞的增殖活性。结果 AB两组纯钛表面细胞密集,胞体丰满,伸展良好,伪足多而长,粗糙表面布满细胞外基质。MTT测得光密度值A组>B组>C组(P<0.05),差异有统计学意义,且每两组间均有差异。结论喷砂粗化双重酸蚀表面处理的纯钛表面有利于人牙龈成纤维细胞的粘附生长且TiO2喷砂处理更优。Objective The purpose of this study was to find a proper surface treatment for soft tissue attachment of human gingival fibroblasts by comparing the effect of three different pure titanium surface treatments in vitro. Methods Pure titanium discs were prepared with 10mm in diameter, lmm thickness and 0.81μm roughness. These discs were divided into 3 groups, group A (TiO2 particle blasting and double acid-etched), group B (Al203 particle blasting and double acid- etched), group C (untreated group). The human gingival fibroblasts were cultured and respectively vaccinated on three group titanium surfaces. The morphology and distribution of human gingival fibroblasts on three group titanium surfaces were observed by scanning electron microscope (SEM). MTT assay were used to measure the proliferative activity of human gingival fibroblasts on three group titanium surfaces. Results Compared with the smooth surface, the attached cells of group A and group B were more significantly, spread well, had longer processes pseudopodia ,and had more extracellular matrix secreted on the slightly rough surface. The results of MTT assay was that integrated optical density of group A was more than that of group B and that of group B was more than that of group C (P〈0.05). The statistic analysis showed that the differences between any two groups were significant. Conclusion Grit-blasting and double acid-etching treatment are profit to adhesion of human gingival fibroblast while Ti02 particles sandblasted and HC1/H2S04 etched treatment is better.
