摘  要:目的:探索2种激光显微切割(laser microdissection)系统应用于切割分离小鼠牙胚特定组织的不同特点。方法:以莱卡LMD6000和蔡司PALM激光显微切割系统分别获取CD1小鼠胚胎期17 d和出生后2 d的下颌第一磨牙牙囊和牙乳头组织。以Trizol法提取2套切割系统获取组织中总RNA,再以RT-PCR法检测两者中管家基因GAPDH表达量的差异。结果:应用莱卡LMD系统获得小鼠下颌第一磨牙牙胚组织中GAPDH基因表达量显著高于以蔡司的PALM系统获得组织中相同基因表达量。莱卡LMD6000激光显微切割系统操作界面友好,切割组织迅速。结论:莱卡LMD6000更适合于下游芯片实验对提取组织RNA完整度较高的要求。Objective: To compare the effects of two types of laser microdissection(LMD)system in isolating the various tissues from murine tooth germ. Methods: The dental follicle and dental papilla tissues of the murine mandibulars first molar at El7 and PN2 sta- ges were dissected by Leica LMD6000 system and Zeiss PALM system respectively. The total RNA was isolated by Trizol method from the dissected tissues and the expression level of house-keeping gene GAPDH from the samples were compared by RT-PCR~ Re- suits : The expression level of GAPDH gene from tissue dissected by Leica LMD6000 system was significantly higher than that by Zeiss PALM system. The operation interface of LMD6000 system was more friendly and its cutting speed was more rapid. Conclusion: Leica LMD6000 system is more suitable for the downstream experiments with higher requirement of total RNA integrity.
