摘 要:目的:通过观察脾虚大鼠马兜铃酸-Ⅰ(AA-Ⅰ)的代谢与肺、肝、胃、小肠组织有机阴离子转运肽2a1(oatp2a1)表达的关系探讨脾主运化的内涵。方法:SD大鼠随机分为空白组、脾虚组、空白+AA-Ⅰ组、脾虚+AA-Ⅰ组。脾虚组、空白组做AA-Ⅰ药代动力学,并取肺、胃、肝、小肠组织;空白+AA-Ⅰ组、脾虚+AA-Ⅰ组予AA-Ⅰ灌胃后5、60m in分别取上述组织,检测各组织中o a tp2a1表达情况。结果:脾虚组与空白组的AA-Ⅰ代谢出现差异。oatp2a1 mRNA表达:脾虚组中肺、肝、胃组织表达量较空白组升高(P<0.01,P<0.05),空白+AA-Ⅰ组60m in中小肠组织表达量较空白组降低(P<0.01),脾虚+A A-Ⅰ组中5min肺、肝组织表达量较脾虚组降低(P<0.05)。结论:脾虚状态下,AA-Ⅰ代谢差异与肺、肝、胃、小肠组织oatp2a1表达改变相关,提示肝、胃、小肠在脾主运化中发挥重要作用,肺发挥协调作用。Objective: To explore the extensive concept of spleen governing transportation and transformation on the relationship between the expression of oatp2al in lung, liver, stomach and small intestine and the metabolism of AA- I in spleen-deficiency rats. Methods: SD rats were randomly divided into 4 groups: blank group, spleen-deficiency group, blank+AA- I group and spleen-deficiency+AA- I group. The pharmacokinetics of AA- I were made and tissues of lung, stomach, liver, small intestine were taken in blank group and spleen-deficiency group; 4 tissues were taken after being given AA- I in 5min and 60min in blank+AA- I group and spleen-deficiency+AA- I group. Expression level of the 4 kinds of tissues in each group was detected. Results: The metabolism of AA- I in blank group and spleen-deficiency group was different. The oatp2al mRNA expressed higher in lung, liver and stomach tissues in spleen-deficiency group compared to blank group (P〈0.01, P〈0.05). The oatp2al mRNA expressed lower in small intestine tissue in blank+AA- I 60min group compared to blank group (P〈0.01). The oatp2al mRNA expressed lower in lung and liver tissues in spleen-deficiency+AA- I 5min group compared to spleen-deficiency group (P〈0.05). Conclusion: The change of the expression of oatp2al is correlated with the metabolism of AA- I in spleen-deficiency rats which suggests that lung, liver, stomach and small intestine might play a great role in the function of spleen governing transportation and transformation.
