摘 要:目的建立当归补血汤中当归的鉴别和阿魏酸的含量测定方法,为当归补血汤质量标准的建立奠定基础。方法采用薄层色谱法定性鉴别当归补血汤中的当归药材,并利用高效液相色谱法测定阿魏酸含量。色谱柱:Diamonsil ODS-C18(5μm,250 mm×4.6 mm)分析柱;流动相:甲醇-0.5%乙酸水溶液(44∶55);流速:1.0 ml·min-1;柱温:30℃;进样量:10μl;检测波长:324 nm。结果当归的薄层色谱鉴别具有较高的专属性,高效液相色谱检测阿魏酸方法具有一定的专属性、准确性、重现性和可行性,样品在0.012 6~0.252μg范围内呈良好的线性关系,平均加样回收率为99.71%,RSD=0.78%。结论所建立方法简便、准确,高效液相色谱分离效果好、线性范围宽、灵敏度高,可用于本品的质量控制。Objective To establish the method for identifying Radix Angelicae Sinensis and detecting the quantity of Ferulic acid in Dangguibuxue Decoction and lay the foundation for the establishment of Dangguibuxue Decoction quality standard. Methods I- dentify Radix Angelicae Sinensis qualitatively by thin layer chromatography (TLC) and detect the quantity of Ferulic acid in Dan- gguibuxue Decoction by High performance liquid chromatography (HPLC). Using Diamonsil ODS - C^s (5 p^m, 250 mm x 4.6 mm) analytical column; the mobile phase was composed of a mixture of methanol - 0.5 % acetic acid solution (44:55 ) ; The flow rate was 1.0 ml ~ rain-~ ;the column temperature was 30~C ;the sample size was 10ul;the detection wavelength was set at 324 nm. Results The TLC method of Radix Angelicae Sinensis is specific. HPLC method for detecting the quantity of Ferulic acid has a good accuracy , reproducibility and feasibility. There was a good linear relationship between the absorption area value and the con- centration in the range of 0. 0126 -0. 252 p^g for sample. The average recovery was 99.71% with the RSD of 0.78%. Conclusion The method is simple and accurate,which has good separation efficiency, wide linear range and high sensitivity. The method can beused to control the quality of Dangguibuxue Decoction.
