摘 要:通过考查茚三酮法、双缩脲法、福林酚法、BCA法、考马斯亮蓝法、凯氏定氮法和TON法测试胶原多肽质量浓度的准确性、稳定性和易操作性,进而评价这些方法用于多肽浓度测试的适用性。实验结果表明,茚三酮法、双缩脲法、福林酚法、BCA法和TON法可以较准确地测定胶原多肽的质量浓度,其中茚三酮法、BCA法和TON法的检测限值低,分别为0.004、0.02、0.02 mg/mL;双缩脲和福林酚法的检测限值较高,为0.3 mg/mL和0.2 mg/mL。茚三酮法、双缩脲法、福林酚法和BCA法的相对标准偏差较低(<6%),表现出较凯氏定氮法和TON法更优的稳定性。此外,茚三酮法和双缩脲法在测试成本和耗时方面具有更强的可操作性。Adaptation of the seven test methods including ninhydrin colorimetry, biuret colorimetry, foline-phenol col- orimetry, bicinch6nininc acid (BCA) colorimetry, coomassie brilliant blue (CBB) colorimetry, kjeldahl and total organic ni- trogen (TON) analysis were used to determine the collagen peptide mass-volume concentration . It evaluates accuracy, re- producibility and operability by these test methods. It was found that collagen peptide mass-volume concentration can be accurately determined by ninhydrin colorimetry, biuret colotimetry, foline-phenol colorimetry, BCA colorimetry and TON analysis, and the lowest detection limits of these five methods are 0.004 mg/mL, 0.3 mg/mL, 0.2 mg/mL, 0.02 mg/mL and 0.02 mg/mL, respectively. The relative standard deviations of ninhydrin colorimetry, biuret colorimetry, foline-phenol col- orimetry and BCA colorimetry are lower than those of kjeldahl and TON analysis, which suggests that these four colorime- try test methods have better reproducibihty. Moreover, ninhydrin colorimetry and biuret colorimetry show the Optimal oper- ability in consideration of test cost and time consuming.
