摘 要:目的探讨柴胡渗湿汤对肺纤维化模型大鼠肺组织中转化生长因子-β1、核因子-κB表达的影响,揭示其治疗肺纤维化的可能作用机理。方法将90只雄性Wistar大鼠随机分成正常对照组、模型对照组、泼尼松组、柴胡渗湿汤组、柴胡渗湿汤+泼尼松组,每组18只。除正常对照组外,其余各组以支气管内注入博莱霉素的方法建立肺纤维化动物模型,于造模后第2天给予相应药物干预;于给药后14 d、28 d在光镜下观察部分大鼠肺组织的病理变化情况,并用免疫组化法检测大鼠肺组织中TGF-β1及NF-κB的表达。结果光镜下模型对照组较正常组肺组织炎症细胞浸润、结构破坏、肺纤维化明显,正常对照组大鼠肺组织TGF-β1和NF-κB的含量较模型对照组显著降低(P<0.05),提示造模成功;分别与柴胡渗湿汤相比,模型对照组肺组织TGF-β1和NF-κB的含量显著增高(P<0.05),泼尼松组两种指标的含量差异无统计学意义(P>0.05),而柴胡渗湿汤+泼尼松组两种指标的含量显著降低(P<0.05)。结论柴胡渗湿汤可能通过抑制肺纤维化大鼠肺组织中TGF-β1及NF-κB的表达,从而延缓大鼠肺纤维化的进展。Objective To discuss the impacts of Chaihu Shenshi Decoction on TGF - β1 and NF - κB in lung tissue of the rats of pulmonary fibrosis and discover the potential mechanism of it in treatment. Methods Ninety male Wistar rats were randomized into a normal control group, a model control group, a Prednisone group, a Chaihu Shenshi Decoction group, and a Prednisone + Chaihu Shenshi Decoction group, 18 rats in each one. Except the normal control group, in the other groups, the bronchial injection of Bleomycin was adopted to set up the model of pulmonary fibrosis. On the 2nd day of modeling, the intervention with corre- sponding medicine was applied. In 14 and 28 days after medication, under microscope, the pathological changes in lung tissues were observed in rats. IHC method was used to detect the expressions of TGF - β1 and NF - κB in lung tissue of rats. Results Under microscope, the results of inflammatory cellular infiltration, struc- tural destroy and puhnonary fibrosis in lung tissue in the model control group were obvious as compared with those in the normal group. The expressions of TGF - β1 and NF -κB in the normal control group were lower significantly than those in the model control group(P 〈0.05) ,indicating the successful modeling. Compared with Chaihu Shenshi Decoction group, separately, the expressions of TGF - β1 and NF -κB were increased significantly as compared with model control group ( P 〈 0.05 ), were not different significantly as compared with Prednisone group(P 〉 0.05 )and were lower significantly than those in Prednisone + Chaihu Shenshi De- coction group ( P 〈 0.05). Conclusion Chaihu Shenshi Decoction delays the progression of pulmonary fibro- sis in the rats possibly through inhibiting the expressions of TGF - β1 and NF - κB in lung tissue.
