利用C6胶质瘤干细胞立体定向建立大鼠脑胶质瘤模型

摘  要:目的利用C6胶质瘤干细胞建立wistar大鼠脑胶质瘤模型,观察肿瘤生长规律及病理特征,探讨利用肿瘤干细胞构建模型的优势。方法体外提取并培养C6细胞系中胶质瘤干细胞,应用立体定向法在鼠脑右侧尾状核区接种104个胶质瘤干细胞,术后连续观察大鼠生存状态和生存时间,分时段行鼠脑MRI检查、病理HE切片及GFAP免疫组织化学检查,观察肿瘤生长及病理特征。结果利用本方法建立模型成瘤率为100%,术后大鼠恢复快,术后反应轻,未见颅外转移,生存期稳定,重复性好,肿瘤生长规律、影像学特征及组织病理特点与人脑胶质瘤相似。结论利用胶质瘤干细胞建立的大鼠脑胶质瘤模型稳定可靠,肿瘤生长更贴近胶质瘤在脑内自然发生的特点,且较传统方法脑内注射体积小,模型制作时间短,术后恢复快,成瘤率更高等优点,为研究胶质瘤干细胞在肿瘤形成过程中的生长方式、分子机制及实验性治疗提供了一个较为理想的模型。Objective To establish a rat brain glioma model in wistar rat with glioma stem cells(GSCs) from C6 gliorna cell line,observe the law of glioma growth, probe into the advantage of establish brain glioma model with GSCs. Methods Separated GSCs from C6 glioma cell line and cultured them in vitro,The GSCs cells (XXXX) were stereotatically implanted into the right caudate nuclei of rats. After inoculation, the living and life span of rats with glioma were observed. The examinations such asMRI, HE staining, immunohistochemical staining of GFAP were performed at different time points after implantation to observe the growth of tumor and the histological charaters. Results Through this method, the rate of tumor formation was 100%. Compared with methods, it shows advantages in postoperative recovery time and complications. The rat brain--glioma model has high reproducibility and stability without extracranial metastasis. The growth characteristics, imaging and pathological features of tumor are similar to human cerebral glioma. Conclusion To establish a rat brain glioma model with GSCs is stable and reliable. The growth of tumor get much closer to the gilioma which occur naturally in human brain. Compared with methods, this method compared with traditional method have some advantages, smaller volume in intracerebral injections,more quickly in establish model, shorter recovery time, higher rate of tumor formation. A rat brain glioma model which established with GSCs can be used as a perfect model to study the growth and molecular mechanism of GSCs in tumor formation and satisfy the need of experimental treatment.
