摘 要:目的分析核迁移蛋白细胞核分离基因C(NudC)与肾癌细胞迁移活性的关系。方法采用Transwell方法比较293和OS-RC-2两个细胞株的迁移能力;RT-PCR和Western blot方法分别检测两个细胞株中NudC mRNA和蛋白的表达差异;免疫荧光方法检测NudC蛋白在上述两个细胞株中的定位。结果肾癌细胞株OS-RC-2的迁移能力明显高于胚肾细胞株293。在mRNA水平,人肾癌细胞株OS-RC-2中NudC表达均高于人胚肾细胞株293(2.6±0.3vs.1.0±0.1)(P<0.05)。免疫荧光结果和细胞核蛋白表达结果显示NudC蛋白在人肾癌细胞株OS-RC-2的核内表达高于人胚肾细胞株293(4.1±0.3vs.1.0±0.2)(P<0.05)。结论 NudC蛋白在肾癌细胞株核内的表达高于正常细胞株,提示NudC可能与肾癌细胞迁移活性密切相关。Objective To investigate the relationship of nuclear distribution gene C(NudC) and the migration activity of renal cancer cells. Methods The Transwell migration assay was applied to observe the migration activity of human embryonic kidney cell line 293 cells and OS-RC-2 cells. RT-PCR and Western blot were used to detect the expressions of NudC mRNA and protein in these cell lines. Immunofluorescence assay was used to detect the localization of NudC protein in 293 and OS-RC-2 cell lines. Results The migration of renal carcinoma cell line OS-RC-2 was higher than human embryonic kidney cell line 293. The mRNA expression of NudC in renal carcinoma cell line OS-RC-2 was higher than that in human embryonic kidney cell line 293 (2. 6 ± 0. 3 vs. 1.0 ± 0. 1) (P〈0. 05). The expression of NudC protein in nucleus was higher in renal carcinoma cell line OS-RC-2 than that in human embryonic kidney cell line 293 (4. 1±0. 3 vs. 1.0± 0. 2)(P〈0. 05). Conclusion The NudC protein expression in nucleus is higher in renal carcinoma cells than that in normal cells suggesting that the expression of NudC may be closely related to the migration activity of renal cancer cells.
