摘  要:目的 观察地塞米松治疗非冻结性冷损伤时神经内膜血管通透性变化,探讨其对血-神经屏障的保护作用.方法 24只雄性Wistar大鼠随机分成低温组与干预组:低温组一侧坐骨神经予以3~5℃持续低温2h;干预组一侧坐骨神经予以相同低温2h,完毕后腹腔注射地塞米松.每只大鼠对侧坐骨神经作为对照.冷损伤后23h,静脉注射伊文思蓝.注射后1h,测量坐骨神经中伊文思蓝吸光度;坐骨神经包埋制片,在荧光显微镜下观察.结果 冷损伤后1d,低温侧坐骨神经中伊文思蓝浓度显著升高,神经内膜中可见伊文思蓝弥漫性分布.给予地塞米松干预后,伊文思蓝浓度明显下降,伊文思蓝几乎局限于神经内膜的血管内.结论 地塞米松治疗坐骨神经非冻结性冷损伤后,神经内膜血管通透性改善,提示可能保护血-神经屏障.Objective To observe the endoneurial blood vessel permeability of sciatic nerves in non-freezing cold injury after treatment of dexametbasone, and discuss protective effect of dexamethasone on the blood-nerve barrier.Methods 24 male Wistar rats were randomly divided into cooled group and intervention group. Sciatic nerves in the cooled group received 3- 5℃ for 2 hours, and sciatic nerves in the intervention group received 3- 5℃ for 2 hours and injected dexamethasone in addition. The other sciatic nerves were the control. After 23h, rats were intravenously injected with Evans blue. Bilateral nerves were harvested lb after the injection. The endoneurial blood vessel permeability were examined by the absorbance measurement and florescence microscope. Results On the I st day, Evans blue concentration in cooled nerves was significantly greater than that in control nerves. Florescence microscopic examination revealed intense red fluorescence in endoneurium, outside the vascular. After treatment, Evans blue concentration decreased significantly, and it showed a bright red fluorescence confined to the lumen of endonenrial blood vessels.Conclusion After non-freezing cold injury, dexamethasone could improve the permeability of endoneurial blood vessel, and may protect blood-nerve barrier.
