胰腺癌细胞 TYMS 基因多态性与蛋白表达的关系

摘　　要：[目的]探讨胸苷酸合成酶（thymidylate synthase ，TYMS）基因多态性与胰腺癌细胞 TYMS蛋白表达的关系。[方法]利用 PCR及凝胶电泳分析7株胰腺癌细胞（AsPC-1，BxPC-3，Capan-1，SU86．86，T3M4， PANC-1与COLO357）TYMS基因型，使用Western Blot检测其 TYMS蛋白表达水平。[结果]7株人胰腺癌细胞TYMS基因5＇-UTR串联重复序列基因分型结果显示形成3种基因型，分别是2R/2R（T3M4与BxPC-3），2R/3R（ASPC-1，Capan-1与 SU86．86）与3R/3R（COLO357与 PANC-1）。不同TYMS基因型蛋白表达OD比值分别为：3R/3R基因型细胞（COLO357与 PANC-1）1．724±0．062，1．063±0．134；2R/2R（T3M4与 BxPC-3）0．558±0．042，0．578±0．021；2R/3R（Capan-1，ASPC-1与 SU86．86）0．530±0．061，0．541±0．0396，0．612±0．036。3R/3R基因型细胞TYMS蛋白表达明显高于2R/2R和2R/3R基因型细胞，两组之间TYMS蛋白表达水平差异有统计学意义（ P ＜0．05），但2R/2R与2R/3R两组之间TYMS蛋白表达无统计学意义（ P ＞0．05）。[结论]在胰腺癌细胞株中，TYMS不同基因型与其蛋白表达密切相关。[Objective]To explore the correlation between gene polymorphism and protein expression of thymidylate synthase（TYMS） in pancreatic cancer cells .[Methods]PCR and gel electrophoresis were used to analyze the genotypes of TYMS in 7 pancreatic cancer cells（AsPC-1 ,BxPC-3 ,Capan-1 ,SU86 .86 ,T3M4 ,PANC-1 and COLO357） .The ex-pression level of TYMS protein in pancreatic carcinoma cells was analyzed by Western Blot .[Results]The result of 5＆#39;-UTR tandem repeat sequence genotyping of TYMS in 7 pancreatic carcinoma cell lines showed 3 genotypes including 2R/2R（T3M4 and BxPC-3） ,2R/3R（ASPC-1 ,Capan-1 and SU86 .86） and 3R/3R（COLO357 and PANC-1） .The optical density（OD） ratio of 3R/3R genotype（COLO357 and PANC-1） of TYMS was 1 .724 ± 0 .062 and 1 .063 ± 0 .134 ,re-spectively .The OD ratio of 2R/2R genotype（T3M4 and BxPC-3） of TYMS was 0 .558 ± 0 .042 and 0 .578 ± 0 .021 ,re-spectively .The OD ratio of 2R/3R genotype（ASPC-1 ,Capan-1 and SU86 .86） of TYMS was 0 .530 ± 0 .061 ,0 .541 ± . 0396 and 0 .612 ± 0 .036 ,respectively .The expression of TYMS in cells with 3R/3R genotype was obviously higher than that in cells with 2R/2R genotype and 2R/3R genotype ,and there was significant difference in the expression of TYMS protein between two groups （ P〈0 .05） ,but there was no significant difference in the expression of TYMS protein be-tween 2R/2R group and 2R/3R group（ P〉0 .05） .[Conclusion]In pancreatic carcinoma cell lines ,different genotypes of TYMS are closely correlated with the expression of TYMS protein .
