热休克蛋白60在左侧和右侧结肠癌中的表达研究

摘　　要：[目的]筛选左侧结肠癌和右侧结肠癌的差异表达蛋白，并确定热休克蛋白60（Heat shock pro-tein60，HSP60）在左侧结肠和右侧结肠癌组织中的差异表达情况。[方法]收集左侧和右侧结肠癌组织及癌旁组织。应用二维电泳、质谱分析和生物信息学分析获得左侧和右侧结肠癌间的差异表达蛋白，并应用 RT-PCR ，Western Blot和免疫组织化学技术检测HSP60在左右侧结肠癌中表达情况，并分析其表达与患者临床病理变量的关系。[结果]建立了左侧和右侧结肠癌的蛋白质组差异表达谱，并发现HSP60是左侧和右侧结肠癌间差异表达蛋白，经过RT-PCR ，Western Blot和免疫组化检测证实 HSP60在左侧结肠癌组织表达强度和表达率显著高于右侧结肠癌，且差异均有显著性（ P ＜0．01）。[结论]HSP60在左侧结肠癌和右侧结肠癌中的表达存在差异，可能在左右侧结肠癌生物学行为差异中有重要意义。[Objective]To screen the differentially expressed protein in left and right side colon cancer ,and to confirm the differential expression of heat shock protein 60（HSP60） in left and right side colon cancer .[Methods]Tissue samples of left and left side colon cancer were collected .The differentially expressed proteins were obtained by using two-dimensional gel electrophoresis（2-DE） ,mass spectrographic analysis and bioinfor-matics .RT-PCR ,Western blot and immunohistochemical method were used to detect the expression of HSP 60 in left and right side colon cancer .The relationship between the expression and clinical pathological variables was analyzed .[Results] The differential expression profiles of proteome of left and right colon cancer were es-tablished .HSP60 was found to be the differentially expressed protein of left and right colon cancer .RT-PCR , Western blot and immunohistochemical method confirmed that the expression intensity and rate of HSP 60 in left side colon cancer were markedly higher than those in right side colon cancer ,and there was significant difference（ P 〈0 .01） .[Conclusion] HSP60 is differentially expressed in left side and right side colon cancer , and may be of important significance in the difference of biological behavior between left side and right side co-lon cancer .
