摘  要:目的:探讨经皮三叉神经电刺激预处理对戊四氮(PTZ)诱发的急性癫痫大鼠的行为学及海马致痫细胞因子白细胞介素1β(IL-1β)及肿瘤坏死因子α(TNF-α)的影响。方法:动物随机分为对照组、致痫组(PTZ组)和经皮三叉神经电刺激组,分别给予7 d、14 d和28 d的假刺激和三叉神经电刺激预处理后,腹腔注射PTZ建立急性癫痫动物模型,观察给药后大鼠癫痫行为学表现,并分别用免疫组织化学方法及ELISA方法对海马IL-1β、TNF-α进行检测。结果:经皮三叉神经电刺激可以明显减轻大鼠的痫性发作级别,减少癫痫发作持续的时间(P<0.05),且海马细胞因子IL-1β及TNF-α的表达明显少于PTZ组(P<0.05或P<0.01)。结论:经皮三叉神经电刺激预处理在PTZ急性点燃癫痫大鼠模型中不仅具有抗惊厥作用,还可以减少海马致痫细胞因子IL-1β及TNF-α的表达,可能为癫痫的防治带来新的策略。AIM: To observe the effect of pretreatment with external trigeminal nerve electrostimulation (eT- NS) on behavioral changes and the expression of interleukin-1β (IL-1β) and tumor necrosis factor α (TNF-α) in hippo- campus of pentylenetetrazol ( PTZ)-treated rats. METHODS : The rats were randomly divided into control group, PTZ group and eTNS group, and kindled by PTZ after administered 7 d, 14 d and 28 d of consecutive fake electrostimulation or eTNS. Subsequently, the severity and duration of seizure were quantitatively evaluated. The concentrations of IL-1β and TNF-α in hippocampus were detected by the methods of ELISA and immunohistochemisty. RESULTS : Compared with PTZ group, treatment with eTNS significantly inhibited the severity and duration of seizure ( P 〈 0.05 ), and significantly re- duced the content of IL-1β and TNF-α in the hippocampus after status epilepticus ( P 〈 0.05 or P 〈 0.01 ). CONCLU- SION: Pretreatment with eTNS may provide a new approach for prevention and treatment of epileptogenesis.
