慢性酒精中毒对清醒大鼠相关脑区部分氨基酸含量变化的影响

摘　　要：目的探讨慢性酒精中毒对清醒大鼠苍白球腹侧核(ventral pallidum,VP)和中脑腹侧被盖区(ventral tegmental area,VTA)细胞外液部分氨基酸类神经递质含量的影响。方法建立慢性酒精中毒自由饮模型采用脑内微量透析法及高效液相色谱法测定清醒自由状态下大鼠苍白球腹侧核和中脑腹侧被盖区细胞外液中的部分氨基酸类神经递质的含量,包括谷氨酸(glutamate,Glu)、天冬氨酸(aspartate,Asp)、谷氨酰胺(glutamine,Gln)、甘氨酸(glycine,Gly)、丙氨酸(alanine,Ala)和牛磺酸(taurine,Tau)。结果苍白球腹侧核区6%酒精组大鼠细胞外液Gly、Tau、Ala含量增高,12%酒精组大鼠细胞外液Gln、Asp含量明显减少。中脑腹侧被盖区6%酒精组大鼠细胞外液Asp、Glu含量均明显增高,12%和8%酒精组大鼠细胞外液Asp、Glu、Ala、Gly、Gln含量均明显增高,而Tau含量在VTA区无明显变化。结论慢性酒精中毒与苍白球腹侧核区Gly、Tau、Ala含量增加有关;慢性酒精中毒与苍白球腹侧核区Gln、Asp含量减少有关;慢性酒精中毒与中脑腹侧被盖区Asp、Glu、Ala、Gly、Gln含量增加有关。Objective To discover the influence of chronic alcoholism on change of partial amino acids neurotrans- mitter content in extracellular fluid of ventral pallidum（VP） and ventral tegmental area（VTA） in healthy rats. Methods Free drinking model of chronic alcoholism was established, the in-brain micro dialysis method and high efficiency liquid chromatography were adopted to detect some amino acids neurotransmitter content in extracellular fluid of ventral pallidum and ventral tegmental area in healthy rats. Result Gly, Tau and Ala content of rats in the extracellular fluid in VP in- creased in 6% alcohol concentration group, and the 12% alcohol group rats showed significant dropping in Gin and Asp content in the extracellular fluid in VP. The 6% alcohol group rats showed significant increase of Asp and Glu content in the extracellular fluid in VTA and the 12% and 8% alcohol group rats showed significant increase in Asp, Glu,Ala, Gly and Gln content in the extracellular fluid in VTA while Tau content had no obvious changes in VTA. Conelusions The chronic alcoholism is related to the increase of Gly,Tau and Ala content in VP. The chronic alcoholism is related to the reduction of Gin and Asp content in VP. The crhonic alcoholism is related to the increase of Asp,Glu,Ala,Gly and Gln content in VTA.
