摘 要:目的观察老年大鼠心肌过氧化氢(H2O2)、丙二醛(MDA)及谷胱甘肽过氧化物酶(GSH-Px)含量的变化及阿托伐他汀干预的影响。方法选取20月龄Wister大鼠90只,随机分为老年对照组、高剂量阿托伐他汀组[10 mg/(kg·d)]和低剂量阿托伐他汀组[1 mg/(kg·d)],每组30只;另选取3月龄Wister大鼠30只作为青年对照组。高、低剂量组每天给予相应剂量阿托伐他汀灌胃,老年对照组给予相同体积的0.9%氯化钠溶液灌胃,青年对照组不做任何处理。灌胃4个月后留取大鼠心肌标本,分别检测各组大鼠心肌H2O2、MDA及GSH-Px。结果老年对照组心肌H2O2、MDA含量均显著高于青年对照组(P<0.01),GSH-Px活性显著低于青年对照组(P<0.01)。高、低剂量组H2O2、MDA含量均显著低于老年对照组(P<0.05,P<0.01),GSH-Px活性显著高于老年对照组(P<0.01);高、低剂量组H2O2、MDA含量均显著高于青年对照组(P<0.05,P<0.01),而GSH-Px活性显著低于青年对照组(P<0.01)。高剂量组上述指标变化较低剂量组更为显著(P<0.05,P<0.01)。结论阿托伐他汀可显著抑制老年大鼠心脏的衰老性改变,可能与其对心脏脂质过氧化水平的抑制有关。Objective To investigate the content changes of H2O2, Malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) in myocardium of elderly rats and the effect of Atorvastatin intervention on it. Methods A total of 90 Wister rats of 20 months old were randomly divided into elderly control group (group A, n = 30) , high dose of Atorv- astatin groupl group B, 10 mg/(kg, d), n=30 and low dose of Atorvastatin group group C, 1 mg/(kg, d), n= 30 ; another 30 Wister rats of 3 months old were chosen as young control group (group D). Rats in group B and C were lavaged with corresponding dose of Atorvastatin every day; rats in group A were lavaged with same volume of 0. 9% sodi- um chloride solution, and nothing was given to group D. After 4 months of intragastric administration, the myocardium samples were obtained, and contents of H2O2, MDA and GSH-Px in the four groups were respectively detected. Results Compared with those in group D, contents of H2 02 and MDA in group A were significantly higher (P 〈 0.01 ) , and GSH-Px liveness was significantly lower (P 〈 0.01 ). Compared with those in group A, the contents of H2O2 and MDA were significantly lower (P 〈 0. 05, P 〈 0. 01 ) , and GSH-Px liveness was significantly higher ( P 〈 0.01 ) ; Compared with those in group D, the contents of H2O2 and MDA were significantly higher(P 〈0.05 ,P 〈0.01 ) , and GSH-Px live- ness was significantly lower (P 〈0.01 ) ; the changes in group B were more significant than those in group C (P 〈0.05, P 〈 0.01). Conclusion Atorvastatin may significantly inhibit the changes of heart aging in elderly rats, which may be related with inhibition to the lipid peroxidation level.
