光活化的金丝桃素诱导胶质瘤细胞调亡的实验研究

摘　　要：目的探讨光活化的金丝桃素对体外培养胶质瘤细胞凋亡的诱导作用。方法体外培养的 Ｗｉｓｔａｒ大鼠 Ｃ６胶质瘤细胞株，加入金丝桃素后进行光诱导作用，然后应用 ＭＴＴ法、３Ｈ－ＴｄＲ掺入实验、ＤＮＡ片断化分析等方法，测试金丝桃素对Ｃ６细胞的细胞毒作用及诱导细胞凋亡作用。结果金丝桃素在低剂量２μｇ／ｍｌ时，７５Ｗ钨灯光照１０ｍｉｎ，即可对Ｃ６细胞株具有细胞毒作用；抽提其ＤＮＡ做琼脂糖凝胶电泳可显示凋亡特有的ＤＮＡ“阶梯”。结论金丝桃素在光诱导下具有抗胶质瘤作用。Objective To determine whether light-induced photoactivation of hypericin can induce apoptosis in malignant glioma in vitro.Methods On the basis of cell culture, C6 cell line was treated with different amotmt of hypericin which was induced by light, then we applied Mrs assay,3H-TdR assay and agarose gel electrophoresis to indicate apoptosis if C6 cells induced by hypericin. Results Hypericin(75w tungsten lamp,10 min of light exposure,2μg/ml)had obvious cytotokic ef- feet on C6 cells, and the specific DNA ladder of apoptosis was displayed. Conclusion light-induced photoactivation of hypericin probably possesses antiglioma effect.
