摘  要:目的探讨Peroxiredoxin(PRDX)Ⅵ mRNA在小鼠卵泡发育和卵巢衰老过程中表达。方法分别选用C57BL/6j小鼠不同周龄、不同动情周期以及促性腺激素预处理卵巢,通过实时定量聚合酶链反应检测PRDXⅥmRNA表达。结果 PRDX Ⅵ mRNA在新生(3D)小鼠卵巢中表达为3周龄组的1.47倍、为6周龄组的3.89倍、为8月龄组的4.28倍(P<0.05);在小鼠动情前期卵巢中表达高于动情后期和动情间期(P<0.05);在促性腺激素预处理试验中,在孕马血清注射48 h后卵巢中表达是对照组的1.7倍,而人绒毛膜促性腺激素注射8、10及12 h后分别为对照组的0.44、0.41、0.54倍,而48 h后则为对照组的2.32倍(P<0.05)。结论 PRDX Ⅵ可能通过调节卵巢氧化应激压力而在卵泡发育和卵巢衰老中发挥重要调控作用。[ Objective ] To explore the expression of Peroxiredoxin Ⅵ mRNA in the different stages of C57BL/6j mouse follicle development and ovarian aging. [Methods] Real-Time Polymerase Chain Reaction was used to de- tect the expression of PRDX Ⅵ mRNA in different stages of age, estrous and Gonadotropin-primed inmature C57BL/ 6j mouse ovaries. [ Results ] Detecting the expression of PRDX Ⅵ mRNA in the ovaries of different age stage of C57BL/6j mice ,we informed that the PRDX Ⅵ mRNA expressed highest in the ovaries of newborn mouse (3D) which were 1.47 times to the 3W group, 3.89 times to the 6W group and 4.28 times to the 8M group (P 〈0.05). Then compared to the diestrus and metaestrus groups, the PRDX Ⅵ mRNA expressed more thickness in the proestrus groups (P 〈0.05). In addition, we noticed that the expression of PRDX Ⅵ mRNA increased notable in mouse ovaries after 48 hours of PMSG injection that was 1.7 times to the control group, and diminished striking as the HCG treated for 8 h, 10 h and 12 h that were 0.44, 0.41, 0.54 to the control group individually, then stepped up to the highest at the 48 h which was 2.32 times to the CON group (P 〈0.05). [Conclusions] The PRDX Ⅵ probably plays important roles in the follicle development and ovarian aging by the regulation of the oxidative stress in the ovary.
