摘 要:目的检测3种Abl相互作用蛋-1(ABI-1)相关的microRNA(miRNA)在胃癌组织及癌旁正常组织中的表达模式,筛选出与胃癌组织相关的靶向ABI-1的miRNA。方法收集59例胃癌术后蜡块标本,包含胃癌组织和癌旁正常组织,抽提石蜡组织中的总RNA。在基因网选择ABI-1相关的miRNA(即miR-181c、miR-148b、miR-9),采用逆转录实时荧光定量PCR法对胃癌组织及癌旁正常组织中的3种miRNA进行检测,计算胃癌组织相对癌旁正常组织中miRNA的相对表达量(2-ΔΔCt)。结果 miR-181c、miR-148b、miR-9在胃癌组织中的相对表达量分别为(2.32±1.04)(P=0.000)、(0.82±0.67)(P=0.321)、(1.07±0.95)(P=0.774),其中miR-181c在胃癌组织及癌旁正常组织中的表达差异有显著性(P=0.000)。结论 miR-181c在胃癌组织中相对高表达,提示其可能作为靶向ABI-1的miRNA调节ABI-1蛋白的表达,为进步研究靶向ABI-1miRNA在胃癌发生发展中的作用奠定基础。[ Objective ] To detect the expression patterns of three kinds of microRNA (miRNA) related to Abelson interactor protein-/ (ABI-I) in gastric carcinoma and adjacent normal tissue, and screen miRNA targeting ABI-1 which may be relevant to gastric carcinoma. [ Methods ] Tissue specimens were from 59 gastric cancer patients who underwent gastrectomy in our hospital, which were nlatched gastric carcinoma and adjacent normal tissue samples. Total RNA in paraffin-embedded tissues were extracted, miRNA (miR-181c, miR-148b, miR-9) relevant to ABI-1 were selected from Gene network. The expression of three miRNAs in gastric carcinoma and adjacent normal tissue were measured using reverse transcriptase real-time quantitative PCR, and the relative expression level (2-am) in gastric cancer tissues were calculated. [ Results ] The relative expression levels of miR-181c, miR-148b and miR-9 in gastric cancer tissues were (2.32±1.04)(P =0.000), (0.82±0.67)(P =0.321), (1.07±0.95)(P =0.774), respectively. The difterenee of expression of miR-181c in gastric carcinoma and adjacent normal tissue was statistically significant (P =0.000). [Conclusions] The relatively high miR-181c expression in gastric cancer tissues suggests that miR-181c maybe target ABI-1 and regulate the ABI-1 protein expression. These results lay the foundation for further study of the role of miRNA targeting ABI-1 in the pathogenesis of gastric cancer.
