精神应激致内脏高敏感大鼠的心理行为学变化

摘　　要：目的观察应用精神应激方法复制内脏高敏感动物模型过程中大鼠的心理行为学变化。方法采用持续避水应激刺激方法复制内脏高敏感动物模型,通过分析结直肠扩张(colorectal distention,CRD)诱发的腹壁收缩反射(AWR)及肌电活动改变对模型进行鉴定,在制模过程中持续观察大鼠体质量、肠道运动(排便颗粒)改变并采用旷场实验观察大鼠的心理行为学变化。结果①应激组的大鼠60 mmHg CRD压力刺激下的AWR评分及VMR与假性应激组和空白对照组相比,差异均有统计学意义;②随着应激天数的增加,应激组的大鼠的体质量逐渐下降,应激前后比较差异有统计学意义(P<0.05),且随着应激天数的增加,排粪粒数逐渐下降,但应激前后比较差异无统计学意义(P=0.172);③应激刺激后大鼠的站立次数及跨越分格线数显著下降,差异有统计学意义(P<0.05)。结论持续避水应激刺激可成功复制内脏高敏感动物模型,显著抑制大鼠体质量增加,改变其肠道动力,降低大鼠兴奋性,诱发其抑郁样行为,为其潜在机制的进一步研究提供实验基础。【Objective】 To observe psychological behavior changes of mental stress induced visceral hy- persensitivity animal model in rats. 【Methods】 Water avoidance stress was given to Wistar rats. To measure rat＇s visceral sensitivity, the abdominal withdrawal reflex（AWR） and the visceral motor response（VMR） to colorectal distension was recorded. And in modeling, the weight, fecal pellet output and behavior changes in open field test were observed. 【Results】 Compared with sham-WA and control group, the AWR score and the mean amplitude of the electromyogram（AUC per second） to 60 mmHg CRD was significantly enhanced in WA group（P 0.05）; With the increase of Water avoidance stress day, the weight and the number of fecal pellet output per hour in WA group rats was decreased, and there is a significant difference（P 0.05） be- tween weight before and after stress, but absent in fecal pellet output（P =0.172）; The number of rats standing up and the number of line crossing were significantly decreased after water avoidance stress（P 0.05）. 【Conclusion】 Water avoidance stress can reduce IBS in rats, inhibits the weight gain, changes the colon motility, reduce the excitability and induced depression-like behavior in rats.
