摘 要:目的探讨海洛因对大鼠胃窦胰多肽(PP)、嗜铬粒素A(CgA)阳性细胞表达的影响,以及PP和CgA在海洛因依赖时的可能作用。方法成年SD大鼠66只,体质量180~220 g,由贵阳医学院实验动物中心提供。按配对原则随机分为正常对照组(n=6)、盐水对照组(n=30)和海洛因依赖组(n=30),处死取胃窦组织,制成连续石蜡切片,采用免疫组织化学SABC法及图像分析技术观察PP和CgA在大鼠胃窦中的表达。结果海洛因依赖组胃窦PP阳性细胞免疫染色强度增强,细胞数量显著高于正常对照组和盐水对照组,平均灰度值低于正常对照组及盐水对照组;CgA阳性细胞的数量及免疫染色强度显著降低,平均灰度值升高,且在第31天时变化最为明显。结论大鼠胃窦PP和CgA的表达受海洛因的影响发生改变,提示这两种细胞通过合成和分泌的变化,参与了海洛因依赖期间肠道功能紊乱的调节过程。【Objective】 To explore the effects of heroin on the expression of pancreatic polypeptide(PP), chromogranin A(CgA) positive cells in gastric antrum of rats,and comprehend the possible functions of them during heroin dependence. 【Methods】 Adult 66 SD rats were randomly divided into three groups: normal control group(n =6), saline control group(n =6) and heroin dependent group(n =30). They were sacrificed and their gastic antrums were taken, embedded in routine paraffin and then made into serial sections, immunohis- tochemical SABC method and image analysis technique were performed to detect the expression of PP and CgA. 【Results】 The number of PP positive cells increased, the intensity of immunohistochemical staining stepped up gradually with the time away during dependence. The results of image analysis showed that the average grey degree of PP positive cells decreased, it is lower than the normal control group(NCG) and saline control group(SCG). The intensity of immunohistochemical staining and the number of CgA positive cells in gastric antrum in heroin dependence group(HDG) dropped obviously compared with in NCG and SCG during heroin dependence, the average grey degree was remarkable higher than that in NCG and SCG, the prominent change appeared at 31 d. 【Conclusion】 Expressions of PP and CgA were changed obviously by heroin, suggesting that they involved in regulation of dysfunction in intestinal tract by changing synthesis and secretion during heroin dependence period.
