摘  要:目的 分析纤维蛋白原(Fg)β-148 C/T基因多态性与性别、年龄、吸烟等因素对血浆Fg水平的影响。方法用PCR-RFLP的方法分析基因多态性,比浊法测定血浆Fg水平。结果病例组血浆Fg7k平明显高于对照组(P<.01),两组的T-148基因频率分布差异无显著性(P>0.05)。男女两性的血浆Fg水平比较差异无显著性(P>0.05):两性中T-148基因携带者血浆Fg水平均比 CC基因型者明显升高(P<0.05)。在 T-148携带者中,血浆 Fg 水平随年龄增长而升高 (P<0.05);吸烟组 Fg 水平高于不吸烟与戒烟组(P<0.05)。结论 Fgβ-148 C/T基因多态性与血浆 Fg水平相关,T-148基因携带者更易受年龄、吸烟等因素影响使血浆Fg水平升高。提示Fgβ-148 C/T基因多态性可作为血栓性疾病的遗传易感标志之一。Objective To analyze the associations of β-fibrinogen gene-148 C/T polymorphism, gender, age and smoking habit with plasma fibrinogen levels. Methods The Fgβ -148 C/R gene polymorphism was analyzed by PCR-RFLP. And plasma fibrinogen levels were determined by Turbimetry,Results Cases was similar in frequency of T-148 allele to con- trol subjects (P>0.05). No significant difference in frequency of T-148 allele was seen in experiment and control pops (P< 0.05).The plasma fibrinogen levels in patients were significantly higher than those in the control subjects(P<0.01). There were no significant differences in plasma fibrinogen levels between men and women (P>0.05); those with T-148 alleles had significant higher plasma fibrinogen levels than those without T-148 alleles(P<0.05). In those with T-148 allele, the Plasma fibrinogen levels were significantly different between the different age groups(P<0.05); smokers had significantly higher plasma fibrinogen levels than non-smokers and ex-smokers (P<0.05). Conclusion Fgβ -148 C/T substitution is a strong and consistent predictor of plasma fibrinogen levels, but the degree of the association is affected by age and smoking, and the T-148 allele may be a susceptible predictor of ischemic stroke.
