摘 要:目的:①探讨具有干细胞标记特性的ALDHCD44+细胞在乳腺癌组织及癌旁组织中的表达情况。②探讨乳腺癌ALDHCI)44+细胞与肿瘤免疫组化指标ER、PR、Her2及Ki67之间的关系。方法:①实验组选取24例乳腺癌患者的术中新鲜癌组织,另取17例癌旁2cm组织标本作为对照。机械一酶方法消化组织、收集细胞悬液,并行肿瘤细胞原代培养,再经流式细胞仪检测ALDHCD4g+细胞的含量。②利用统计学相关分析及£检验方法了解乳腺癌原代细胞中ALDHhiCD44‘细胞与临床免疫组化指标ER、PR、Her2、Ki67之间相关性。结果:①24例浸润性乳腺癌组织中有22例原代培养成功(培养成功率为91.67%),17例癌旁组织中有14例能够培养出原代细胞(培养成功率为82.35%)。②ALDHhiCIM4+细胞在癌组织原代培养的细胞中含量为(4.56±2.92)%(0.56%-10.30%),在癌旁组织原代细胞中含量为(0.15±0.06)%(0.05%-0.23%),两值相比差异有统计学意义(P〈0.05)。@ALDHhiCD44‘细胞与ER(P=0.065)、PR(P=0.107)、Her2(P=0.050)表达量之间差异无统计学意义,无相关性,与Ki67(P=0.007)表达量之间差异存在统计学意义,相关系数r=0.593。结论:①利用肿瘤组织原代细胞培养、检测方法,进一步证实ALDHhiCD44+乳腺癌干细胞的存在,其在癌灶的表达量高于癌旁组织。②结合患者临床病理结果分析,显示乳腺癌干细胞ALDHhiCD44+与Ki67之间存在相关性,与ER、PR、Her2指标之间无相关性。Objective: To explore the expression of ALDHh breast cancer stem cells in breast cancer tissues and cancer ad jacent tissues, and explore the relationship between ALDHhi CIM4 + cells and ER, PR, Her2, Ki67. Methods: The fresh breast cancer specimens of 24 cases during surgery were collected as experimental group and the cancer adjacent tissue specimens of 17 cases were collect- ed as control group ; the specimens were digested by mechanical - enzyme method, cell suspension was collected to perform primary culture, and then the content of ALDHhi CIM4 + cells were detected by flow cytometry ; the relationship between ALDHhl CD44 ~ cells and ER, PR, Her2, Ki67 was analyzed with t test. Results: Among 24 cases of invasive breast cancer tissues, primary culture was successful in 22 cases, the successful rate was 91.67% ; among 17 cases of cancer adjacent tissues, primary culture was successful in 14 cases, the successful rate was 82. 35% ; the contents of ALDHhl CD44 + cells in invasive breast cancer tissues and cancer adjacent tissues were (4. 56 ± 2. 92 ) % (0. 56% - 10. 30% ) and (0. 15± 0. 06 ) % ( 0. 05 % - 0. 23 % ), respectively, there was statistically significant difference ( P 〈 0. 05 ) ; there was no correlation between ALDHhiCIM4 cells and ER (P =0. 065) , PR (P =0. 107) , and Her2 (P =0. 050) ; but there was a sig- nificant correlation between ALDHhi CD44 cells and Ki67 expression ( P = 0. 007, r = 0. 593 ) . Conclusion : The exitance of ALDHhi CIM4 cells were further confirmed through primary culture and detection of breast cancer cells, and its expression level in breast cancer tis- sues was higher than that in cancer adjacent tissues; the study showed that there was a correlation between ALDHhiCD44 cells and Ki67, but there was no correlation between ALDHhiCD44 cells and ER, PR, Her2.
