摘　　要：目的检测WWOX、p53及Bcl-2mRNA及其蛋白在鼻咽癌(NPC)组织中的表达情况,并分析其在鼻咽癌中的作用。方法用RT-PCR和免疫组织化学染色方法检测61例鼻咽癌和20例鼻咽黏膜慢性炎症(CIONM)组织中WWOX、p53及Bcl-2mRNA及其蛋白表达情况,并探讨其意义。结果 WWOX mRNA及其蛋白在鼻咽癌组织中的表达低于鼻咽黏膜慢性炎症组织,WWOX蛋白表达与鼻咽癌不同的临床分期及有无淋巴结转移有关。在鼻咽癌中,p53和Bcl-2mRNA及其蛋白呈高表达,p53蛋白的表达在鼻咽癌晚期明显高于早期,WWOX蛋白的表达与p53和Bcl-2表达呈负相关关系。结论在鼻咽癌组织中,WWOX、p53和Bcl-2存在异常表达,且WWOX与p53和Bcl-2蛋白的表达呈负相关关系,WWOX在鼻咽癌的低表达可能与该病的发生、发展及转移有关。Objective To detect the mRNA and protein expressions of WWOX,p53 and Bcl-2 in nasopharyngeal carcinoma （NPC）tissues and to examine their role in the development of NPC. Methods The expressions of WWOX,p53 and Bcl-2 were detected in 61 cases of NPC and 20 cases of chronic inflammation of nasopharyngeal mucosa（CIONM）by RT-PCR and immuno- histochemistry. Results The mRNA and protein expression of WWOX was significantly lower in NPC tissues than in CIONM tissues. The protein expression of WWOX was associated with clinical stages and lymph node metastasis of NPC. p53 and Bcl-2 were found overexpressed in NPC,and the protein expression of p53 was much stronger in advanced stage NPC than in early stage. Moreover,it was found that the expression of WWOX was negatively correlated with the expressions of p53 and Bel- 2. Conclusion WWOX,p53 and Bcl-2 are abnormally expressed in NPC with negative correlation found between the expression of WWOX and the expressions of p53 and Bcl-2. The down-regulation of WWOX may be involved in the occurrence, progression and metastasis of NPC.
