摘 要:目的研究华蟾素对膀胱癌T24细胞的体外抑制作用,探讨其对膀胱癌的临床应用价值。方法用MTT法测定不同浓度华蟾素和配伍丝裂霉素C(MMC)作用下的T24细胞24h、48h的抑制作用;用流式细胞术(flowcytometer,FCM)测定不同浓度华蟾素作用T24细胞48h后细胞周期表现。结果(1)华蟾素组:25、2.5、0.25、0.025、0.001mg/ml华蟾素作用T24细胞24h后,生长抑制率分别为52.7%、47.2%、38.9%、41.7%、33.3%;作用48h后,分别为58.3%、50.0%、47.4%、47.4%、44.8%。MMC组:0.001、0.0005mg/mlMMC作用24h后,抑制率分别为71.1%、60.6%;48h后,分别为75.0%、72.2%。配伍组:0.001mg/ml MMC与2.5mg/ml华蟾素、0.0005mg/ml MMC与2.5mg/ml华蟾素,作用24h后抑制率分别为80.6%、68.4%,两组间差异有统计学意义(P<0.05);48h后抑制率分别为84.2%、72.2%。与MMC组比较,相同MMC浓度下配伍组24h、48h抑制率差异均有统计学意义(P<0.05)。(2)0.025、0.25、2.5mg/ml华蟾素作用T24细胞48h后发现T24细胞G0/G1期分别占76.3%、78.7%、79.6%。结论华蟾素对体外T24细胞有明显抑制作用,且呈时间及浓度依赖性;具有G0/G1期阻滞作用;与MMC有协同作用;可作为中晚期膀胱癌药物治疗的一种选择。Objective To study the value of cinobufacini injection on clinical application, by observing the inhibitory effects of it on the proliferation of bladder carcinoma cell T24 in vitro. Methods By MTT assay, inhibitory effects were determined at 24,48 hours when T24 cell had been cultured with different concentration of cinobufacini injection and MMC; By FCM, cell cycle of bladder carcinoma cell T24 were determined at 48 hours when they had been cultured with different concentration of cinobufacini injection. Results (1)when T24 cells had been cultured with different concentration of cinobufacini injection: 25,2.5,0.25,0.025,0.001 mg/ml, its 24h growth inhibiting ratio were 52.7%, 47.2%, 38.9%, 41.7%, 33.3% respectively,its 48h growth inhibiting ratio were 58.3%, 50.0%,47.4% ,47.4%,44.8% respectively, and there were some dose-effect and time-effect relationship.when T24 cells had been cultured with 0.001mg/ml,0.000 5mg/mlMMC,its 24h growth inhibiting ratio were 71.1% ,60.6% respectively, its 48h growth inhibiting ratio were 75.0%, 72.2% respectively, and there were obvious dose-effect and time-effect relationship.when T24 cells had been cultured with 2.5mg/ml cinobufacini injection and 0.001,0.000 5mg/ml MMC,its 24h growth inhibiting ratio were 80.6% ,68.4% respectively, its 48h growth inhibiting ratio were 84.2% ,72.2% respectively, and there were obvious dose-effect and time-effect relationship;Compared with MMC team, cinobufacini injection have synergistic effect with MMC. (2)cell cycle change of T24 cell by FCM: when T24 cells had been cultured with 2.5,0.25,0.025mg/ml cinobufacini injection after 48h,we found the cell number during G0/G1 phase were 79.6% ,78.7% ,76.3% respectively. Conclusions cinobufacini injection can inhibit the proliferation of bladder carcinoma cell T24 and arrest the cell cycle during G0/G1 phase in vitro, and show time and concentration dependent;h have synergistic effect with MMC; It can be used to treat bladder carcinoma at advanced stage.
