摘 要:以产油酵母菌Y1为供试菌株,通过单因素及正交试验对其产脂发酵培养基进行优化。结果表明,最优培养基组成为葡萄糖85 g/L、蛋白胨40 g/L、(NH4)2SO410.00 g/L、MgSO4·7H2O 0.300 g/L、Na2HPO43.0 g/L、KH2PO42.5 g/L。按照营养成分最优组合,100 mL三角瓶装液量30 mL,液体种子接种量10%,摇床转速160 r/min,温度28℃,得到的产油酵母菌Y1生物量为20.25 g/L,生物量比优化前提高了46.5%。Using the oleaginous yeast Y1 as the experiment strain,single factor experiments and orthogonal experiment of liquid fermentation medium were optimized.The optimal medium were glucose 85 g/L,peptone 40g/L,(NH4)2SO4 10.00 g/L,MgSO4·7H2O 0.300 g/L,Na2HPO4 3.0 g/L,KH2PO4 2.5 g/L.According to the optimum combination of nutritional components,liquid volume in 100 mL flask was 30 mL,seed inoculation of liquid was 10%,shaking speed was 160 r/min,temperature was 28 ℃,the cell biomass of oleaginous yeast Y1 was 20.25 g/L,the cell biomass increased by 46.5% after the optimization.
