摘 要:目的:优化土茯苓多糖的超声提取工艺,并研究其抗氧化活性。方法:在单因素试验基础上,利用响应面法,以多糖提取率为响应值,建立数学模型,获得最佳工艺;采用总的抗氧化活性和还原能力2种体外抗氧化实验模型初步评价土茯苓多糖的抗氧化活性。结果:通过二次回归模型响应面分析得出土茯苓多糖的最佳提取工艺条件是液固比为35∶1,超声功率90w,提取时间为18.60min,提取2次,在此最佳工艺条件下土茯苓多糖提取率为0.3106%,理论值为0.3255%,与理论值相对误差小于5%。抗氧化活性研究中,土茯苓多糖总的抗氧化活性和还原能力均随多糖浓度的增加而上升。结论:由响应面法优化得到的土茯苓多糖的提取工艺方便可行,得到的土茯苓多糖具有较强的抗氧化活性,值得进一步开发利用。Objective: The optimum conditions of ultrasonic extraction and antioxidant activities of Smilax glabra polysaccharides (SGP) were investigated in this paper. Methods: Response surface method (RSM) was used to establish the mathematical model of ultrasonic extraction of SGP to obtain the optimum conditions based on single factor experiments. The antioxidant activities of SGP in vitro were studied by the methods of reducing power and ORAC assay. Results: The optimum conditions obtained through RSM analysis were as follows: the water/solid ratio 35:1, ultrasonic power 90w, extraction time 18.60min. Under the optimal condition, the predicted value of two-time extraction was 0.3255%, while the experimental value was 0.3106%. The experimental value agreed with the predicted from the regression model with a relative error less than 5%. The total antioxidant activity and reducing Power increased with the increasing concentration of polysaccharides. Conclusion: Optimization of the extraction process by RSM of SGP is convenient and feasible. SGP has good antioxidant activity and is worth to develop for application.
