摘  要:目的研究Wnt5a对肝星状细胞(nsc)促炎细胞因子IL-1β及IL-6表达的影响,探讨Wnt5a在肝纤维化中的作用及机制。方法采用脂多糖(LPS)和肿瘤坏死因子(TNF-α)刺激人肝星状细胞株LX-2,以qRT—PCR和Western blot检测Wnt5a表达水平变化;分别利用Wnt5a重组蛋白体外刺激LX-2和Wnt5a shRNA病毒转染LX-2,以qRT-PCR、ELISA检测促炎细胞因子IL-1β、IL-6的水平变化。结果LPS及TNF-β可显著刺激LX-2 Wnt5a表达增加(P〈0.05);Wnt5a重组蛋白可促进LX-2细胞IL-1β、IL-6表达和分泌(P〈0.05);Wnt5a shRNA转染LX-2后IL-1β、IL-6表达水平显著降低(P〈0.05),ELISA发现Wnt5ashRNA转染组培养上清IL-1β、IL-6水平显著低于pLVX-NC—shRNA组与LX-2组(P〈0.05)。结论Wnt5a可促进HSC促炎细胞因子IL-1β、IL-6的表达而参与肝纤维化进程。[ Abstract ] Objective To study the effects of Wnt5a on the expression of proinflammatory cyto- kines IL-1β and IL-6 in human hepatic stellate cells and its pathogenic mechanism in liver fibrosis. Meth- ods The expression of WntSa at mRNA and protein levels in LPS-stimulated and TNF-α-stimulated human hepatic stellate cell line LX-2 were analyzed by quantitative real-time PCR and Western blot. LX-2 cells were treated with human recombinant Wnt5a protein and transfected by lentivirus carrying Wnt5a shRNA, respectively. The expression of proinflammatory cytokines IL-1β and IL-6 in treated cells were examined by qRT-PCR and ELISA. Results LPS and TNF-α enhanced the expression of WntSa in LX-2 cells (P〈 0.05). Recombinant Wnt5a protein significantly increased the expression and secretion of IL-1β and IL-6 (P〈0.05). Compared with pLVX-NC-shRNA cells and LX-2 cells, the expression and secretion of IL-1β and IL-6 in pLVX-Wnt5a-shRNA transfected LX-2 cells were significantly decreased (P〈0.05). Conclu- sion WntSa might participate in the progression of hver fibrosis through promoting the expression of proin- flammatory cytokines IL-1β and IL-6 in hepatic stellate cells.
